| Rice is an important food crop in China.In the ripening stage of rice,when it encounters high temperature and high humidity environmental conditions,rice ears are prone to panicle germination.The occurrence of panicle germination has a certain impact on the realization of the goal of high yield and high quality of rice in China.In this study,192 indica rice materials were verified by whole ear germination method,and genome-wide association analysis was performed by 40 K SNP chip data,and sites significantly related to the germination resistance of rice panicles were excavated and candidate genes were identified By comparing the candidate gene sequences of panicle-tolerant and non-panicle-tolerant materials,the possible genomic information of functional genes was explored,laying the foundation for the next step of building a functional verification vector and developing molecular auxiliary markers.The main findings of the study are as follows:(1)10 extreme materials were identified as panicle germination resources,and three panicle-resistant germination materials were screened out,including H119,H169,H124;(2)The panicle germination rate of 192 materials in the 0th d after maturation was calculated,and the Q+K hybrid linear model was used,and 40 K SNP was used The chip performs genome-wide association analysis.The results showed that a total of 39 were detected between the 12008078 and 12299303 regions of chromosome 10 SNP loci were associated with panicle-tolerant germination traits,and the phenotypic contribution rate was between 0.07469 and 0.13503;(3)Gene scans were performed within the target region to locate 42 gene products: 9for unknown functional expression,6 hypothetical proteins,5 transposon proteins,and 5transposon proteins for reverse transcription There are 8,14 with well-defined functional homologous proteins;of which 2 are cytochrome P450 proteins.Obtain genetic information through NCBI,predict the function of candidate genes,and speculate LOC_Os10g23130(Os10g0378000),LOC_Os10g23180(Os10g0378100)are most likely to be involved in the genetic regulation of panicle germination tolerance in rice.At the same time,the genome and protein sequence information of the predicted gene are analyzed online,and the target gene of the LOC_Os10g23180 gene is speculated according to the expression value of RNA-Seq FPKM;(4)The promoter sequence analysis of 10 extreme materials showed that the promoters of 10 parts of the material can be divided into two types: type I is missing from-1144 to-1127 bp,including Yi Sui Meng Material m3,H208,H180,H43;type II in-485~-473 bp deletion,including spike-resistant materials H119,H169,H124 and easy-to-panicle materials H62,H61,H52. |
