| Hind limb paralysis is a common clinical disease in pets.The affected dogs and cats are manifested as loss of hind limb motor ability and decreased ability of autonomous control of urine and feces.If it cannot be improved,it will bring great trouble to pet owners in raising and nursing.At present,the common causes of hind limb paralysis include car accident trauma,intervertebral disc herniation,central neuritis and so on,its essence is the injury of the spinal cord.TCM believes that hind limb paralysis caused by spinal cord injury belongs to Bi syndrome and impotence syndrome,which is essentially Qi and blood stasis.Acupuncture stimulation can promote Yang qi recovery and promote Qi and blood operation.The high consistency between the spinal cord and the governor vein,whether in the modern anatomical position or the meridian strike distribution,suggests that there is a close relationship between the two.Electroacupuncture stimulation of the governor vein may solve the hind limb movement disorder caused by spinal cord injury.At present,there are many studies on acupuncture points for spinal cord injury,but there are few studies on Baihui point,which is unique to animals.However,the clinical application of this point is very wide.In this experiment,this special point is selected,combined with Dazhui point for electroacupuncture stimulation,to explore the efficacy and possible mechanism of these two points on the Du vein on spinal cord injury paralysis model.The main research results are as follows:1.Establishment of Allen’s model and postoperative careIn the present study,the spinal cord injury model is the most common one established by the method of heavy weight drop,which has good flexibility and flexibility.In this study,the spinal cord of rats was damaged by the self-made Allen’s percussion apparatus through surgical operation,and the model of hind limb paralysis was established.After the operation,antibiotics were given to prevent infection,and the rats could urinate by themselves until the bladder compliance recovered.2.Effects of electroacupuncture at Dazhui and Baihui points on motor function,survival rate,local blood circulation and morphology of spinal cord tissue in rats with spinal cord injuryAfter the modeling was successful,the rats in the electroacupuncture group were given acupuncture treatment at Dazhui and Baihui points with 7 days of treatment cycle(6 days of acupuncture and 1 day of rest),and 3 cycles of acupuncture treatment were 21 days.The behavioral scores of rats in each group and the results of the inclined plate test were observed and recorded during acupuncture.On the 14 th day of acupuncture,the behavioral scores of rats in the electroacupuncture group and the inclined plate test were significantly different from those in the model group(P(27)0.05),suggesting that the motor function of hind limbs had recovered to some extent.The survival rate and weight change rate of rats in each group were monitored in the experiment(P(27)0.05).It was found that the survival rate of electroacupuncture group was significantly increased,and the weight loss rate was significantly lower than that of the model group(P(27)0.05).During acupuncture treatment,the local temperature changes of acupoints before and after acupuncture were recorded by red Fu radiation trace map.It was found that after acupuncture,the area of high temperature near Dazhui point was significantly expanded,and the temperature of Dazhui point and Baihui point was significantly increased.At the end of the experiment,the spinal cord tissues of rats in each group were taken for HE staining and Nishi staining.The results showed that the number of minor spinal necrosis and Nishi corbosome in the electroacupuncture group was significantly higher than that in the model group(P(27)0.05),suggesting that electroacupuncture may protect the damaged spinal cord and alleviate secondary spinal cord injury.3.Effect of electroacupuncture at Dazhui and Baihui points on serum inflammatory factors and oxidative stress factors in rats with spinal cord injuryModern studies believe that inflammation is the key to secondary injury after spinal cord injury,accompanied by oxygen free radical injury.In this study,ELISA was used to detect the changes of inflammatory factors in serum and spinal cord tissue of rats in each group.The results showed that the content of inflammatory promoting factors in serum of electroacupuncture group was significantly lower than that of model group(P(27)0.01),and the content of inflammatory inhibitory factor IL-10 was significantly higher than that of model group,suggesting that electroacupuncture may avoid further injury of spinal cord tissue by inhibiting inflammatory response(P(27)0.01).Compared with model group,SOD content and MDA content in serum of electroacupuncture group were significantly increased(P(27)0.01),suggesting that electroacupuncture may protect spinal cord tissue by inhibiting oxygen free radical damage4.Effect of electroacupuncture at Dahui and Baihui points on cell apoptosis in rats with spinal cord injuryApoptosis is a kind of programmed cell death,and the initiation mechanism of the program is very complex.In rats with spinal cord injury,apoptosis appears early and persists,which is an important factor affecting the outcome.In this experiment,Bax expression in the model group was significantly up-regulated on the 21 st day of acupuncture treatment(P(27)0.05),suggesting that it was related to spinal cord injury.The expression of Bcl-2 in the electroacupuncture group was significantly up-regulated(P(27)0.05),suggesting that acupuncture at Dazhui and Baihui points may regulate the expression of Bcl family genes to up-regulate the expression of Bcl-2 in the treatment of spinal cord injury and restore spinal cord function.5.Effect of electroacupuncture at Dazhui and Baihui points on nerve growth factors in rats with spinal cord injuryNerve growth factor is involved in regulating the survival,differentiation and functional expression of nerve cells,and has neurotrophic effect on spinal motor neurons,which can promote their functional expression.In this experiment,BDNF expression in spinal tissue of rats in electroacupuncture group was significantly up-regulated after 21 days of acupuncture at Dazhui and Baihui points(P(27)0.05).It is suggested that 21 days of electroacupuncture may promote the repair of spinal cord injury by increasing the content of brain-derived neurotrophic factor.Through this experiment,it was found that electroacupuncture at Dazhui and Baihui points had significant clinical efficacy in the treatment of post-spinal cord injury paralysis model of rats,and the motor function of hind limbs of rats in the electroacupuncture group recovered fastest in the second cycle of acupuncture treatment.After the treatment,the serum inflammatory factors,oxidative stress factors and apoptosis genes of rats in each group were measured.It was found that electroacupuncture at Dazhui and Baihui point could reduce spinal cord injury,protect neurons and promote the recovery of hind limb motor function by inhibiting inflammation,reducing oxygen free radical damage and regulating apoptosis. |
