Objective:Establishing the rat model of chronic reflux esophagitis by" cardiomyotomy combine with pylorus ligation",and connect with cervical vagotomy.To observe the changes of acetylcholine transferase(ChAT),nitric oxide synthase(NOS)activity and cytokines in plasma and esophageal tissues of rats,and to explore the possible mechanism of the effect of Xuanfu Daizhe Decoction on cholinergic pathway of reflux esophagitis.Methods:65 male SD rats were randomly divided into:normal group(10),RE(10)and sham operation group(10),model group(15),traditional Chinese medicine group(10),western medicine group(10).In addition to the normal group,rats in each group were treated with " cardiomyotomy combine with pylorus ligation "to establish the reflux esophagitis model,model group,treatment group were implemented left cervical vagotomy at the same time,the rats in sham operation group were only separated the left vagus nerve.Three weeks after molding,the rats were lavaged for medicine,traditional Chinese medicine group for xuanfu daizhe decoction,western medicine group for mosapride suspension liquid,the other groups were given the same amount of saline.Treated for two consecutive weeks,the rats were sacrificed,and the esophageal tissue and plasma were collected for analysis.Observed the pathological morphological changes of each rats esophageal tissue.Using chemical method to detect NOS activity in esophageal tissue and plasma and using ELISA method to detect the content of ChAT,IL-1 beta,IL-6,IL-8,IL-10,INF alpha in esophageal tissue and plasma.Results:1.Three weeks later,the model group appeared reflux esophagitis,the NOS activity in plasma was significantly lower than that in normal group.The level of ChAT in plasma(P<0.01),the IL-1 beta,IL-6,IL-8 and TNF alpha in esophageal tissue and plasma were significantly higher than those in normal group(P<0.05).2.Compared with the model group,the esophageal mucosa lesion was improved in the traditional Chinese medicine group,and the content of ChAT in plasma of the traditional Chinese medicine increased,but the activity ofNOS did not change significantly.The levels of IL-1beta,IL-6 and TNF alpha in esophageal tissue and the levels of IL-6,IL-8 in plasma were significantly decreased(P<0.05),but the content of IL-10 did not change significantly(P>0.05).3.Compared with the western medicine group,the esophageal mucosa in the traditional Chinese medicine group have no obvious difference,the content of ChAT in plasma was obviously higher than the western medicine group(P<0.05),other index were no statistical difference(P>0.05).Conclusion:Xuanfu daizhe decoction can inhibit rats’ reflux esophagitis esophageal mucosal inflammation,its mechanism may be through increasing the content of ChAT,activating the cholinergic anti-inflammatory pathway,inhibiting IL-1 beta,IL-6,IL-8 and TNF alpha,to improve the esophageal sphincter function and promote restoration of the esophageal mucosa.
