| Objective:1、the establishment of kidney qi deficiency asthma rat model;2、To evaluate the efficacy and possible mechanism of Th17/Treg imbalance in traditional Chinese medicine compound combined with western medicine in rats with renal qi deficiency by flow cytometry.To explore the therapeutic mechanism of prescription Yishen Chuanning Decoction on kidney qi deficiency and asthma,and to provide reliable experimental basis for clinical drug use.Methods:1、48 male Wistar male rats were randomly divided into 6 groups.The model of kidney qi deficiency was simulated by multivariate method.2、After 14 days of continuous modeling,the corresponding drugs were given to the treatment group,and the other groups were treated with normal saline.3、The levels of serum testosterone(T),cortisol(COR)and thyroxine(T4)were measured by ELISA.4、HE staining was performed on rat lung tissue.5、The proportion of Treg and Th17 cells in the spleen of asthmatic rats was measured by flow cytometry,and the correlation between them and airway inflammation was analyzed.Results:1、The tracheal and alveolar morphology of normal rats showed normal,and the lungs showed no inflammation;Asthma group and kidney qi deficiency asthma group,there is inflammation of the lungs,alveolar wall and trachea wall was thickened2、After treatment with ovalbumin sensitization and multivariate model,the values of T4,COR and T were decreased(P<0.05);After treatment,the T4,COR and T values of each drug group increased(P<0.05).3、The changes of Treg and Th17 cells in each group were observed.The percentage of Treg and Th17 cells in the spleen of six groups was analyzed by flow cytometry.Compared with the normal control group,the proportion of Treg cells in the spleen of asthma group and asthmatic group was significantly decreased.Chinese medicine,western medicine,Chinese and western combination group,the change trend was basically the same,compared with the model group,the proportion of cells increased,but still lower than the normal group.While the percentage of Th17 cells in the model group was higher than that in the normal control group,The proportion of Th17 cells in the treatment group was lower than that in the model group,but still higher than that in the normal group.There were Th17/Treg imbalance in the asthma group,th17/Treg ratio was significantly higher than the control group.Conclusions:1、This experiment successfully established asthma and kidney qi deficiency asthma model.2、The levels of T、T4 and COR in the model of kidney qi deficiency were descending.Proved that there is a correlation between the deficiency of kidney qi and T、T4 and COR.3、The proportion of Treg cells in the spleen of the model group was significantly lower than that of the normal group.After treatment,the number of Treg cells in the treatment group increased in different degrees,which in turn increased the most significant combination of Chinese and Western groups.At the same time,the number of Th17 cells in the model group increased,and the proportion of Th17 cells in the treatment group decreased.Suggesting that Treg and Th17 are involved in the pathogenesis of asthma,and Yishenchuanning Decoction and the role of Pumicilu in the treatment of asthma may be associated with Treg,Th17.By enhancing the function of Treg cells,increasing the number of Treg cells,thereby inhibiting the function and number of Th17 cells,reducing the Th17 cell response function,to achieve the purpose of relieving symptoms of asthma.At the same time,the effect of Chinese and western medicine on the promotion of Treg cells and the inhibitory effect on Th17 cells was the most obvious,indicating that the effect of Chinese and Western medicine in the treatment of asthma was the most obvious. |
