Celosia cristata,also known as cockscomb comb head,chicken flowers,for Celosia plumosagroup plant.Because the inflorescence shape and comb similar,so the name cockscomb,enjoy "flowers in the bird" in the world.The plant has a colorful inflorescence and can be widely cultivated throughout the world as an ornamental plant.The main chemical constituents of celosia cristata are flavonoids,steroids,saponins,terpenes.Cockscomb has the effect of hemostasis and convergence,and it is a common folk medicine for gynecology.Commonly used in the clinical treatment of functional uterine bleeding,uterine bleeding,uterine bleeding and other symptoms.Base on the material research and the initial pharmacodynamics research of the material extracted by ethanol,it proves that the materials extr-acted from Celosia cristata by ethanol and ethyl acet-ate have a certain degree of stop bleeding effect.In this study,the chemical constituents of ethyl acetate extract from Cockscomb were isolated and identified.Therefore,the hemostatic mechanism of the extract from Celosia cristata by n-butanol would be studied further.1.Research on chemical composition extracted by ethyl aceyl acetateSeparation was performed on the extracted fraction of ethyl acetate in a 70%ethanol extract of Celosia.The macroporous resin method was used to preliminarily separate the ethyl acetate extract from Celosia.The mixture was purified by normal phase silica gel column chromatography,ODS column chromatography,recrystallization and HPLC(preparative)methods.The structure of the compound was identified by physicochemical constants and spectroscopic techniques.As a result,six monomer compounds were isolated,and the structures of the respective monomer compounds were identified by using spectral data such as 1H-NMR and 13C-NMR spectrometry in the spectrum analysis and measuring the physicochemical constants of the individual monomer compounds.They were finally identified as ferulate-6O-β-D-glucopyranoside(1),quercetin-3-O-β-D-glucopyranoside(2),and valine methyl ester(3),(8S)-7,8,9-trihydric alcohol(4),decagon-3-O-glucoside(5),2-methoxy-4-(2-propenyl)-phenyl-β-D-glucopyran oside(6).The above compounds were isolated from the plants of the genus feather cockscomb for the first time.2.Research on the hemostatic mechanism of extracted by n-butanolMice was given oral administration of mifepristone and misoprostol in early pregnancy rats,resulting in incomplete abortion to establish dysfunctional uterine bleeding model.60 female SD rats were randomly divided into six groups:normal control group,model group,n-butanol effective fractions of Celosia officinalis high,middle,low dose group and Gongxuening group.The hemostatic effect of celosia on functional uterine bleeding was evaluated by the amount of uterine bleeding.It was found that celosia significantly reduced the amount of uterine bleeding.Through the uterine organ coefficient and uterine and ovarian pathological detection,the mechanism of the effect of Celosia Cristata on uterine bleeding after medical abortion was investigated.The NO,NOS and ET values in the uterine homogenate of each group were measured by ELISA.Compared with the normal group,the contents of NO,NOS and ET in the uterine tissue homogenate of the model group were significantly increased(P<0.01).Compared with the model group,the levels of NO,NOS and ET in the uterine tissue homogenate of the uterine blood group were decreased,and the NOS and ET values were statistically significant(P<0.01).Compared with the model group,the high,medium and low dose groups of cockscomb could decrease the contents of NO,NOS and ET in the uterine tissue homogenate,there was significant difference between the high dose group and the model group(P<0.01).Compared with Gongxuening group,the high dose group of cockscomb was better than Gongxuening group.Cockscomb by regulating the uterine tissue homogenate in the NO,NOS,ET levels,regulation of uterine tissue NO,NOS,ET balance,to promote endometrial repair,uterine vasoconstriction,so as to achieve the purpose of bleeding.
