Research purposes:The paper discusses the recombinant lentivirus mediated hFGF21 transfer to type 2 diabetic rats,it ability to regulate glucose metabolism and its related signaling pathways.Experimental method:(1)Construction of recombinant plasmid containing hFGF21 gene,and by lentivirus packaging becoming lentivirus particle.Type 2 diabetic rats were estabilshed by high fat diet and STZ.(2)The recombinant lentiviral vector was injected into the liver of type 2 diabetic rats,and the expression of exogenous hFGF21 gene in liver of type 2 diabetic rats was detected.(2)After the recombinant lentiviral plasmid was transferred into type 2 diabetic rats.(3)The fasting blood glucose,ITT,GTT and blood lipid levels in diabetic rats.Detection of the pathological status of rats with type 2 diabetes.(4)Detection of the pathological status of rats with type 2 diabetes(5)Detection of glucose metabolism and its Related protein in type 2 diabetic rats(6)Detection of oxidative stress,apoptosis and related proteins in rats with type 2 diabetes mellitus.Experimental result:(1)Recombinant lentiviral vector carrying hFGF21 gene.(2)After recombinant lentivirus mediated hFGF21 was introduced into type 2 diabetic rats,the expression of exogenous hFGF21 gene and protein was detected.(3)In type 2 diabetic rats,the level of blood glucose was decreased to normal level after the introduction of recombinant lentivirus mediated hFGF21.(4)After hFGF21 treatment of type 2 diabetic rats liver glycogen and muscle glycogen synthesis ability enhancement,and sugar metabolism related proteins IRS1,IRS2,PI3K expression and GSK-3 beta increased significantly.(5)After hFGF21 treatment of type 2 diabetic rats,oxidative stress levels returned to normal range,cell apoptosis significantly reduced the expression of apoptosis related protein NF kappa B and Caspase-3 decreased.Empirical conclusion:FGF21 was able to restore fasting glucose and lipid levels in most T2DM animal models,The results showed that FGF21 can restore the level of glucose metabolism by activating the PI3K/AKT signaling pathway and inhibiting the NF kappa B signaling pathway.
