Objective: To perform the isolation and identification of endophytic fungi of Ningguo Peucedanum praeruptorum Dunn,a genuine medicinal material in Anhui,and its distribution diversity and biodiversity were discussed.The effects of different fermentation methods on secondary metabolites of endophytic fungi were further explored,looking for compounds with novel structures and diverse activities.Methods:The classical tissue block method was used to isolate the endophytic fungi of Ningguo P.praeruptorum Dunn,and morphology and molecular biology were used for the identification of these fungi.The diversity and community distribution of endophytic fungi in Ningguo P.praeruptorum Dunn were analyzed by statistical method.The chemical constituents of fermentation products of Two Endophytic Fungi of Ningguo P.praeruptorum Dunn were studied by modern chromatography and spectroscopy.The obtained monomer compounds were tested for cytotoxicity.Results: 361 endophytic fungi were isolated from Ningguo P.praeruptorum Dunn and identified as 31 genera and 62 species.Through the Sorenson similarity index and cluster analysis,it can be concluded that the similarity of endophytic fungi in Ningguo P.praeruptorum Dunn in three different regions is relatively low,with obvious differences.Fusarium tricinctum and Aspergillus udagawa are the dominant species.Two endophytic fungi,A.udagawae and F.tricinctum,were fermented in two different fermentation modes to obtain the secondary metabolite ethyl acetate extraction site,and 40 compounds were isolated,of which A.udagawae solid fermentation product was acetic acid.13 compounds were isolated from the ester site,of which 4 were new compounds.F.tricinctum,its the solid fermentation product and the ethyl acetate fraction of the liquid fermentation product were separated into 14 and 13 compounds,respectively.The activity test results of 5 oral cancer cells showed cyclo(D-pro-L-leu)(28),cyclo(S-pro-S-leu)(29),cyclo-(D-pro-L-phe)(37),cyclo-(trans-4-hydroxy-L-prolyl-L-phenylalanine)(40)have good activity.Conclusions: There were no significant differences in the dominant populations of endophytic fungi from P.praeruptorum Dunn in three different regions,and the similarity was low,showing a certain regional specificity;40 monomer compounds were isolated from the fermentation products of 2 endophytic fungi.Compounds 1-4are novel compounds in which the activity of the compounds 28,29,37,40 have good activity.
