Objective:To probe the relationship between tumor-associated macrophages(TAMs)and clinicopathological characteristics and prognosis of DLBCL(Diffuse Large B Cell Lymphoma,DLBCL).Methods:Immunohistochemical method was used to detect the expression of CD68 and CD163 in the tumor tissues of 95 DLBCL patients in the People’s Hospital of Guangxi Zhuang Autonomous Region,and collect the relevant clinical characteristic data of the patients,analyze the expression of CD68 and CD163 in DLBCL,and its clinical characteristics in DLBCL patients,also the relationship between DLBCL patients and its impact on the prognosis of DLBCL patients.Search and download DLBCL related data sets GSE31312,GSE53786,GSE10846 including relevant clinicopathological characteristics and prognostic information from Gene Expression Omnibus(GEO)online.Obtain the M2 macrophage infiltration level in the data set through the Xcell web version,analyze the relationship between the M2 macrophage infiltration group and the clinicopathological characteristics and prognosis of the patient,and the subpopulation of each infiltrating immune cell in the M2 macrophage infiltration group Infiltration level.Results:There are 1098 diffuse large B-cell lymphoma patients were contained in the study,including 95 patients from the People’s Hospital of Guangxi Zhuang Autonomous Region,and probably 1003 patients in the three data sets of GSE10846,GSE31312,and GSE53786.The results showed that high expression rates of CD68 and CD163 in diffuse large B-cell lymphoma were47.3% and 35.7% respectively.T-test analysis results indicated that the expression of CD68 was significantly different in different groups of Ann Arbor staging,and CD163 was expressed in gender,age,B symptoms,ECOG PS,LDH level,β2 microglobulin,Ann Arbor staging,whether there were large lumps and bone marrow invasion There are significant differences in the expression of different groups.Univariate survival analysis testify that the expression of CD68 and CD163 are the prognostic factors that affect the DLBCL patients’ progression free survival(PFS).In the GEO data set,T-test analysis of M2 type macrophages has a even higher level of infiltration in the ABC subtypes patients,ECOG PS score <2 and older than 60 years old.Univariate survival analysis showed that the level of M2 macrophage infiltration is a factor affecting overall survival(OS),and patients which has low M2 macrophage infiltration have a better prognosis.Multivariate survival analysis results showed that the high expression of M2 macrophage infiltration is an independent risk factor which influence the prognosis of DLBCL.The level of infiltrating immune cells in M2 macrophages in the high and low infiltration group showed that NK cells,NKT cells,CD8+ T cells,neutrophils,basophils and have high infiltration levels in the M2 macrophage high infiltration group.The infiltration level of B cells and mast cells was high in the low infiltration group of M2 macrophages.Conclussions:1.The high expression of CD68 and CD163 is a bad factor affecting the prognosis of DLBCL.2.In the DLBCL GEO data set,the level of M2 macrophage infiltration is concerned with the patient’s age,pathological subtype,and ECOG PS score.3.The infiltration level of tumor infiltrating immune cell subsets is relevant to the infiltration level of M2 macrophages.4.High level of M2 macrophage infiltration is an independent risk factor affecting the prognosis of DLBCL.
