Objective: 1 Based on the study of ancient and modern medical books and the experience of doctors in the application of panax notoginseng,this paper systematically evaluates the clinical application of panax notoginseng in intestinal diseases.2 To explore the relationship between the angiogenesis-related proteins(VEGF and Ang-2)and angiogenesis,verifying the anti-angiogenic utility of Panax notoginseng.From the point of view of anti angiogenesis,it can provide a new idea for the prevention and treatment of colitis.3 To study the action mechanism of Panax notoginseng in treating ulcerative colitis by using the network pharmacology.Methods: 1.1 Ancient literature research Refer to the ancient and modern classics related to panax notoginseng prescription to understand the historical evolution and changes of the origin,efficacy and main treatment of panax notoginseng prescription.At the same time,refer to the application of panax notoginseng prescription in the treatment of intestinal diseases by ancient doctors and carry out relevant induction and summary.1.2 Modern literature research Search the relevant literature on the application of panax notoginseng in intestinal diseases: the author takes "UC","ulcerative colitis","inflammatory bowel disease","panax notoginseng" and "Tianqi" as the key words,and searches Wanfang database,Chinese Journal Full-text Database,Chinese science and Technology Journal Full-text Database and Pub Med database.Collect literatures on panax notoginseng prescriptions related to the treatment of intestinal diseases.2 Choose the clean-grade male kunming mice and the mouse animal model of ulcerative colitis could be constructed by DSS free drinking method.Different drugs were respectively infused for 1 week,observing and recording the general condition and disease activity index(DAI)score of the UC mouse.After drawing materials,doing the hematoxylin-eosin staining method to observe the histopathological changes;conducting the immunohistochemical examination to detect the expression level of VEGF and Ang-2 in UC tissue and normal colon tissues and organize the data.3.1 Obtaine the active components and targets of notoginseng by TCMSP and other TCM compound databases combining with literature miningand.Obtain the target of Panax notoginseng in the treatment of ulcerative colitis by the Databases of Gene Cards,OMIM and DRUGBANK.3.2 The "Panax notoginseng-component-target" network was plotted with Cytoscape software.The PPI network was constructed with Cytoscape software and STRING database to obtain the core action target of Panax notoginseng for ulcerative colitis.3.3 Conduct the GO analysis and KEGG pathway analysis of targets of notoginseng for UC by using the Metascape platform tool.Results: 1 The general condition change,the disease activity index(DAI) score and the pathological histology suggested the successful preparation of the ulcerative colitis model.Light microscope suggested that the model group had defect or shedding necrosis,ulcer formation,recess structure disappearance,abscess formation,infiltration of inflammatory cells in the layer propria;the treatment group showed significant improvement compared with the model group,but the colon mucosa was complete,but there were mucosal defects,crypt abscess,some bleeding points,and more inflammatory cells infiltrated the lamina propria.Positive rate of VEGF and Ang-2 and average optical varied in normal mice,model group and each drug intervention group.VEGF,Ang-2 are important indicators of angiogenesis,with positive brown-yellow staining,VEGF and Ang-2 were expressed at high levels in the model group,in the tissues of each drug intervention group,and their positive rate was significantly higher than that in normal colonic tissues(P<0.05);Compared with the model group,the reduced protein expression levels in each drug intervention group could improve the UC symptoms to varying degrees(P<0.05).The results suggested reduced angiogenesis in each drug intervention group.The results suggested reduced angiogenesis in each drug intervention group.2.1 The screen yielded 8 chemical components of Panax notoginseng,190 corresponding targets,1924 of ulcerative colitis,and 119 ginseng components and disease common targets.2.2 Several targets of the top-ranked key proteins in the PPI network are AKT1,JUN,TNF,PTGS2,and CASP3.2.3 The GO and KEGG enrichment analysis results yielded 1584 biological processes,142 molecular functions,61 cell components and 187 signaling pathways;KEGG analysis of UC treatment involves the main targets of IL1 A,VEGFC,RB1,ANGPT1,and ANGPT2,and the regulated pathways are mainly related to signaling pathways such as PI3K-Akt,p53,NF-kappa B,and VEGF.Conclusions: 1 Panax notoginseng can improve the colon damage and inflammatory response of UC mice induced by DSS in UC mice to some extent,which may be related to the effective downregulation of VEGF and Ang-2 protein expression and the inhibition of pathological angiogenesis,so as to play the role of prevention and treatment of UC.2 The treatment of UC is probably closely related to VEGFC and ANGPT2,and its mechanism of action is related to the regulation of signaling pathways such as PI3K-Akt,p53,NF-kappa B,and VEGF.
