| Objective: The effects of LncRNA RP11-502I4.3 on the proliferation,migration and angiogenesis of human retinal microvascular endothelial cells(HRMECs)were investigated under hyperglycemic environment in order to provide experimental basis for exploring the molecular mechanism of LncRNA RP11-502I4.3 in the occurrence and development of DR.Methods: 1.qRT-PCR was used to detect the expression of LncRNA RP 11-502I4.3and VEGF in HRMECs under normal(5.5mmol/L)and high glucose(30.0mmol/L)conditions;2.Lentivirus transmission was used to overexpression of LncRNA RP11-502I4.3,then,HRMECs were divided into the following four groups: Normal Glucose group(NG),High Glucose group(HG),High Glucose + overexpressed negative control(HG+ oe-NC),High Glucose +LncRNA P11-502I4.3 overexpression group(HG+ oe-RP11-502I4.3),qRT-PCR was used to detect the expression of LncRNA RP11-502I4.3 and VEGF in HRMECs of each group.3.The proliferation ability of each group was detected by CCK-8 method;4.Transwell assay was used to detect the migration ability of cells in each group;5.The tube formation ability of each group was analyzed by tube formation experiment.Results: 1.qRT-PCR analysis showed that compared with NG group,in HG group,the expression of LncRNA RP11-502I4.3 in HRMECs was decreased,while the expression of VEGF was increased,the difference was statistically significant(P<0.05).2.Compared with HG group,VEGF expression in HRMECs was down-regulated after overexpression of LncRNA RP11-502I4.3 under high glucose environment,and the difference was statistically significant(P<0.05).3.CCK-8 cell proliferation results showed that: Compared with NG group,the proliferation activity of HRMECs in HG group was increased,and the difference was statistically significant(P<0.05).Compared with HG group,the proliferation activity of HRMECs in overexpressed LncRNA RP11-502I4.3 group was significantly decreased(P<0.05),while there was no significant difference in overexpressed control group(P>0.05).4.Transwell test results of cell migration showed that: Compared with NG group,the number of HRMECs migration in HG group was significantly increased,and the difference was statistically significant(P<0.05).Compared with HG group,the number of HRMECs migration in LncRNA RP11-502I4.3 group was significantly decreased(P<0.05),while there was no significant difference in the overexpression control group(P>0.05).5.Analysis of angiogenesis results of HRMECs by tube formation experiment showed that: Compared with NG group,the formation of tube was significantly increased in HG group,and the difference was statistically significant(P<0.05).Compared with HG group,the formation of tube was significantly decreased in LncRNA RP11-502I4.3 group(P<0.05),while there was no significant difference in the overexpression control group(P>0.05).Conclusions: LncRNA RP11-502I4.3 may inhibit the proliferation,migration and tube formation of HRMECs induced by high glucose. |
