| Background and purpose:The purpose of this study was to investigate whether CBLB could inhibit the migration and invasion by inhibiting the epithelial-mesenchymal transition in osteosarcoma cells,which provides a theoretical basis for targeted therapy of osteosarcoma.Method After transient transfection up-regulated or knockdown CBLB expression,the ability of proliferating,migration and invasion were tested by CCK-8,wound-healing and Transwell assays.Next,changes of EMT-related proteins were detected after CBLB expression in up-regulated or down-regulated cells.TGF-β was added into upregulated CBLB cells,and its effects on migration and invasion were detected by Transwell and scratch assay.Results:1.After up-regulating the expression of CBLB in osteosarcoma cells,the results of CCK-8 showed that the proliferation ability of osteosarcoma cells was obviously inhibited and scratch test and Transwell test showed that the migration and invasion ability of overexpressed CBLB cells were lower than that of control group;2.After down-regulation the expression of CBLB in osteosarcoma cells,its inva sion and migration was promoted;3.In CBLB overexpression cells,EMT related protein E-cadherin expression w as increased,N-cadherin and Vimentin protein were decreased,but E-cadherin and Vimentin protein expression were increased after CBLB down-regulation;Moreover,TGF-β could attenuate the migration and invasion of osteosarcoma cells after up-regulation of CBLB。Conclusion:CBLB can inhibit EMT and decrease the invasion and migration of osteosarcoma cell.CBLB may be a underlying biomarker for treatment of osteosarcoma. |
