Objective:To analyze the expression level of lncRNA TUG1 in middle ear cholesteatoma tissues and its correlation with clinical stage of middle ear cholesteatoma patients and damage degree of ossicles.Methods:The expression level of lncRNA TUG1 in middle ear cholestatoma tissues was detected by real-time fluorescence quantification(QRT-PCR).Pearson and Spearman correlation coefficients were used to analyze the correlation between TUG1 and clinical stage and damage degree of ossicles in patients with middle ear cholesteatoma.Results:TUG1 expression was increased in cholesteatoma tissues of middle ear patients(P<0.001),which was correlated with clinical stage(P<0.001).The expression of TUG1 in middle ear cholesteatoma tissues increased(P<0.001).Pearson correlation coefficient showed that TUG1 was positively correlated with ossicular destruction score(R =0.709,P<0.001).In the prognostic follow-up,the expression level of TUG1 in recurrent choleadiatoma tissues was also higher than that in non-recurrent choleadiatoma tissues.The sensitivity and specificity of TUG1 in predicting 1-year recurrence of patients was 81.82% and 86.00%(P<0.001).Conclusion:In this study,it was found that the increased expression level of TUG1 in middle ear cholesteatoma tissues was correlated with clinical stage,and was positively correlated with the degree of destruction of patients’ ossicle,and with disease recurrence one year later,suggesting that TUG1 may be a new indicator for the diagnosis and treatment of middle ear cholesteatoma in the future.
