Effects Of Jiaji-electroacupuncture Combined With Self-made Rehabilitation Exercise Running Wheel On Motor Function And Oxidative Stress Injury In Rats With Spinal Cord Injury

Objective:To observe the effects of Jiaji electroacupuncture combined with self-made rehabilitation exercise running wheel on the improvement of motor function,the spinal cord tissue morphological changes and the expressions of LPO,MDA and nNOS in the injured area of spinal cord injury,to explore the Jiaji electroacupuncture combined with self-made rehabilitation exercise running wheel method to inhibit oxidative stress injury,reduce the expression of nNOS protein,and promote motor function recovery and the regeneration of nerve cells in rats.Methods:72 healthy adult female SD rats aged ten weeks,weighing 200±20g.The rats were randomly divided into four groups,with 18 rats in each group:sham operation group,model group,Jiaji electroacupuncture treatment group,Jiaji electroacupuncture combined with self-made rehabilitation exercise running wheel group.The corresponding treatment in Jiaji electroacupuncture treatment group and Jiaji electroacupuncture combined with self-made rehabilitation exercise running wheel group was carried out on the third day after successful modeling,and the rats were in stable condition.The treatment was last for 28 days.The BBB score was collected after treatment on the 7th,14th and 28th day,and in each group were randomly selected 6 rats.The injured local tissue was taken for paraffin section,HE staining observed the morphological changes of spinal cord tissue,immunohistochemical method observed the positive expression of nNOS,and Elisa method observed the positive expression of LPO and MDA.Results:(1)BBB score:After SCI,the BBB score of the sham operation group was 21,the BBB score of the other three groups were the lowest on the 7th day,increased on the 14th day,and the highest on the 28th day.At the same time point,the BBB score of the Jiaji electroacupuncture group and the Jiaji electroacupuncture combined with self-made rehabilitation exercise running wheel group was lower than that of the sham operation group(P<0.05),higher than that of the model group(P<0.05),and the score of the self-made rehabilitation exercise running wheel group was higher than that of the Jiaji el ectroacupuncture group(P<0.05).(2)Enzyme linked immunosorbent assay(Elisa):At 7d,14d and 28d after SCI,the positive expressions of LPO and MDA in all groups(except sham operation group)decreased with time,reaching the highest on the 7th day,then gradually decreased and the lowest on the 28th day,while the positive expressions of LPO and MDA in the two treatment groups were higher than those in the sham operation group(P<0.05)and lower than those in the model group(P<0.05)at the same time point.It was lower in Jiaji electroacupuncture combined with self-made rehabilitation exercise running wheel group than that in Jiaji electroacupuncture group(P<0.05).(3)Results of immunohistochemical detection:The results of immunohistochemistry showed that the positive expression of nNOS protein in all groups(except sham operation group)decreased with the extension of time at 7 days,14 days and 28 days after SCI.The positive expression of nNOS in the two treatment groups was higher than that in the sham operation group(P<0.05)and lower than that in the model group(P<0.05)at the same time point,while that in the Jiaji electroacupuncture combined with self-made rehabilitation exercise running wheel group was lower than that in the Jiaji electroacupuncture group(P<0.05).(4)Observation local injured spinal cord in SCI rats of histomorphological changes by eosin staining:In Sham operation group,the tissue structure was intact and the morphology of neuronal glial cells was no abnormality seen;In the model group,the spinal cord tissue was seriously deformed and lost,neurons were lost,and a large number of inflammatory cells were infiltrated;After treatment,the number of neurons in Jiaji electroacupuncture group and Jiaji electroacupuncture combined with self-made rehabilitation exercise running wheel group increased,the number of inflammatory cells decreased,and the neurons were repaired to some extent;After treatment,the nerve cells in Jiaji electroacupuncture combined with self-made rehabilitation exercise running wheel group were neatly arranged and the repair effect of nerve cells was more obvious than that in Jiaji electroacupuncture group.Conclusion:1.Both Jiaji electroacupuncture and Jiaji electroacupuncture combined with self-made rehabilitation exercise running wheel can increase the BBB score of spinal cord injury rats,improve spinal cord tissue structure,and reduce the expression of LPO,MDA and nNOS in local spinal cord injury.Jiaji electroacupuncture combined with self-made rehabilitation exercise running wheel treatment is more significant than Jiaji electroacupuncture.2 Jiaji electroacupuncture combined with self-made rehabilitation exercise wheel therapy can promote nerve cell regeneration and improve motor function in spinal cord injury area of rats,which may be related to the inhibition of LPO,MDA and nNOS protein expression.
