| Objective: To investigate the expression of KIF20A and CDCA7 protein in patients with non-small cell lung cancer(NSCLC),and to analyze the relationship among KIF20A and CDCA7 protein expression and clinicopathological characteristics and prognosis in NSCLC patients.Materials and Methods: To select 126 cases of NSCLC and 126 cases corresponding adjacent lung tissues(>1cm)by surgical resection between January2018 and June 2021 from the pathology department of affiliated hospital of You Jiang Medical University for Nationalities.Their clinicopathological data were collected.Using immune histochemical technique to detect the expression of KIF20A,CDCA7 and Ki67 in 126 NSCLC and lung tissues adjacent.50 NSCLC patients were detected for EGFR gene mutation by PCR-fluorescence probe method randomly.Using SPSS13.0 and statistical methods to analyze the relationship between KIF20A,CDCA7,Ki67 and EGFR gene mutations and clinical and pathological features of NSCLC patients.The correlation and prognosis among KIF20A,CDCA7expression and EGFR gene mutation in NSCLC was analyzed by statistical methods.Results:(1)KIF20A and CDCA7 proteins were not expression or low expression in adjacent lung tissues,but significantly increased in NSCLC tissues.And it was statistically different(P<0.001).(2)The expression of KIF20A in NSCLC was related to the degree of clinical staging,tumor differentiation,lymph node metastasis,Ki67 and airway spread(P<0.05).And there was no link among tumor size,gender,age,histopathological type,smoking,distant metastasis and pleural invasion(P>0.05).(3)CDCA7 expression in NSCLC was correlated with clinical staging,lymphatic metastasis,Ki67,distant metastasis and airway spread(P<0.05),but has no relationship with gender,age,tumor size,tumor tissue type,differentiation degree of tumor,pleural invasion and smoking(P>0.05).(4)The expression of KIF20A has on any correlation with CDCA7(r=0.062,P=0.493).(5)The detection rate of EGFR gene mutation rate in 50 NSCLC patients was 58% and exons 19 and 21 accounted for the most.It was not associated with KIF20A and CDCA7 expression and clinicopathological data(P>0.05).(6)According to the Kaplan-Meier survival curve and Log-rank described by the experimental results,the overall survival time of patients who were high expression of KIF20A was significantly shortened and the prognosis was worse(P=0.002).There was no significant difference in overall survival between patients with high CDCA7 expression and those with low expression(P=0.170).(7)Cox proportional hazard regression showed that KIF20A were risk factor for the prognosis of NSCLC patients(P=0.034).Conclusion: Through this project for the master’s degree,we found that the expression of KIF20A and CDCA7 protein was upregulated in the tumor tissues from patients with NSCLC.The upreguation of KIF20A and CDCA7 protein was associated with tumor proliferation,clinical stage,lymphatic metastasis,and airway spread.KIF20A expressionwas an independent risk factor affecting the prognosis of NSCLC patients. |
