| Objective:This study confirmed the lipid-lowering effect of portulaca oleracea by animal experiment.Furthermore,the lipid-lowering mechanism of Portulaca oleracea was further discussed by using network pharmacology,which provided scientific theoretical basis for clinical nurses to use portulaca oleracea to regulate the diet of patients with hyperlipidemia,and also laid a foundation for further research work.Methods:1.Fifty SPF ICR male mice were randomly divided into 5 groups according to body weight,10 mice in each group,namely blank group,model control group,fenofibrate group,purslane water decoction low-dose group,purslane water decoction high-dose group.Except blank group,the mice in the other four groups were modeled by fat emulsion intragastric administration.After successful modeling,the mice were given intragastric administration for 21 days.Blood samples were collected from bilateral eyeballs of mice 1h after the last administration,and the blood lipid indexes were detected by automatic biochemical analyzer.Liver weight was measured and organ index was calculated.During the whole experiment,the normal water intake of mice in each group was guaranteed and the intake of ordinary feed was recorded.2.Based on the results of animal experiments,the pharmacologic analysis was carried out on the network.Firstly,the active components of Portulaca oleracea were screened through TCMSP database.Swiss Target Prediction database was used to predict the effective chemical constituents of Portulaca oleracea.On the other hand,hl P-related targets were screened by Gene Gards,OMIM,Drug Bank and TTD disease databases.The intersection of disease target and component target was obtained by Venny platform,and the data of intersection target was imported into STRING database for protein interaction network analysis,and the target protein interaction network was constructed by Cytoscape3.8.2software.Finally,GO analysis and KEGG analysis of target genes were conducted by Metascape online database.Results:1.Effect of PORTulaca oleracea l.on blood lipid level in mice: Compared with model group,CHO(cholesterol),TG(triglyceride),LDL(low density lipoprotein)and glucose indexes of portulaca oleracea l.high-dose group were decreased(P<0.05);The level of HDL(high density lipoprotein)was increased(P<0.05),and the difference was statistically significant.Compared with model group,TP(total protein),ALT(alanine aminotransferase)and AST(aspartate aminotransferase)levels in PORTulaca oleracea high-dose group were decreased(P<0.05).However,ALB(albumin)level increased(P<0.05).2.Effect of PORTulaca oleracea on liver: Compared with model group,the liver weight and liver organ index of mice in portulaca oleracea low-dose group and high-dose group were decreased to a certain extent(P<0.05).3.Effect of Portulaca oleracea on body weight of mice: Compared with the model group,the body weight of mice after administration was decreased to a certain extent,and the body weight of mice in portulaca oleracea high-dose group at week 5 had statistical significance(P<0.05).4.Effect of Portulaca oleracea on appetite of mice: compared with blank group,the intake of mice in model group decreased to a certain extent,and statistical analysis was conducted(P>0.05).However,the data at week 5 showed that the food intake of portulaca oleracea(low)and portulaca oleracea(high)groups was significantly higher than that of model group,suggesting that portulaca oleracea has a certain effect on appetite promotion.5.Network pharmacology study of Portulaca oleracea in the treatment of hyperlipidemia: a total of 10 active components and 217 drug targets were screened out;A total of 435 hl P-related disease targets were obtained by searching Gene Gards,OMIM,Drug Bank and TTD databases.A total of 55 intersection targets were obtained by mapping drug targets to diseases.The PPI network map of drug and disease targets was constructed,and 20 core targets for HLP treatment were screened out,including TNF、IL6、AKT1、IL1B、VEGFA and TP53.By GO analysis.KEGG analysis revealed 30 enrichment pathways.Conclusion:1.Portulaca oleracea can effectively reduce the body weight,liver weight and liver index of hyperlipidemia model mice,and can protect the liver of mice;2.Portulaca oleracea can effectively reduce the lipid index of model mice,increase the food intake and promote appetite.3.This research from the aspect of network pharmacology preliminary reveals the horse tooth yellow prevention mechanism of action of HLP,predict possible dent by lipid and atherosclerosis,fluid shear stress and sclerosis of arterial from sample,diabetes complications of AGE-RAGE signal path,way,TNH signal pathways in cancer signaling pathways play a roe of reducing blood fat,for further in-depth experimental Research provides evidence. |
