| In this paper,the freshly picked peony petals in Heyang,Shaanxi Province,were freeze-dried,and in order to improve their utilization,the wall breaking rate of the lyophilized very fine powder of peony petals was detected by microcounting method;the nutrient composition of the lyophilized very fine powder of peony petals was analyzed and determined,and the content of 6 chemical components in the lyophilized very fine powder of peony petals was determined by HPLC method;because it mainly contained phenols,flavonoids and other components,a mouse acute and chronic alcoholic liver injury model was established for further development and application.The effect of lyophilized very fine powder on alcoholic liver injury of peony petals was evaluated.Experiment 1: Preparation of lyophilized very fine powder of peony petals and detection of cell wall breaking rate and its nutrient content.Fresh peony petals are quick-frozen(-18 °C)after picking,coarsely crushed,freeze dryer dried,crushed,sifted through 80 mesh to obtain fine powder,and then super finely crushed under conditions below 0 °C,through300 mesh sieve,to obtain peony petals freeze-dried very fine powder(10 °C below conditions preservation).The cell breaking rate of the lyophilized very fine powder of peony petals was measured by the cell counting method,and the cell breaking rate of the lyophilized very fine powder of peony petals was measured by the cell counting method,and the nutritional content was detected in accordance with the relevant experimental methods and requirements of the national food safety standards of the Ministry of Health of the People’s Republic of China.The results showed that the peony petals were freeze-dried and crushed to obtain a very fine powder,and the cell wall breaking rate was measured to reach 100%,which was rich in protein,vitamin C,essential amino acids and chemical elements of the human body and other nutrients.Experiment 2: The content of 6 chemical components in the lyophilized ultrafine powder of peony petals was detected.Peony petals lyophilized very fine powder added to the appropriate amount of ultrapure water,stirred evenly to disperse and dissolve,ultrasonic extraction for 20 minutes,standing,filtered with a microporous membrane,in accordance with the established HPLC method,the sample was detected.The results showed that the contents of gallic acid,hydroxyponyl glycosides,paeonosides,wild sumac glycosides,aperioglobin and apigenin-7-O-β-D-glucoside were between 4.29-5.00 mg/g,2.13-3.32mg/g,1.27-1.81 mg/g,2.92-3.75 mg/g,7.14-7.80 mg/g and 3.12-3.75 mg/g,respectively.Experiment 3: The effect of lyophilized very fine powder of peony petals in the prevention and treatment of acute alcoholic liver injury in mice.The 80 mice were randomly divided into 8 groups of control,model,biphenyl diester,peony petal lyophilized ultra-fine powder in different doses,etc.,with 10 mice in each group.The control group and the model group mice were gastricly filled with equal volume distilled water,and the remaining groups were gastrically filled with 150 mg/kg biphenyl diesters and 20 mg/kg,40 mg/kg,80mg/kg,160 mg/kg,and 320 mg/kg peony petals lyophilized ultra-fine powder once a day for 10 consecutive days.At 30 min after the last administration,except for the control group of mice,the remaining groups had a 56-degree Red Star Er pot head liquor(referred to as white wine)with a volume fraction of 15 m L/kg.The results showed that the content of AST and ALT in the serum of each group of mice with lyophilized very fine powder of peony petals was significantly reduced and showed a certain dose effect,that is,the content decreased more obviously with the increase of the dose;the liver and stomach coefficients were basically at normal levels;the content of TG and MDA in the liver tissue was significantly lower than that of the model group,while the content of ADH and SOD was significantly higher than that of the model group;the content of GSH-PX and CAT in the gastric tissue increased,and the CK and LDH in the heart tissue decreased;the GSH content in the brain tissue was increased,and the NO content decreased;the liver,Observation of histopathological slices such as stomach,heart and brain confirmed that the lyophilized extremely fine powder groups of peony petals had significant changes in edema and inflammation compared with the model group.It shows that the lyophilized ultra-fine powder of peony petals has obvious effects on the prevention and treatment of acute liver and stomach damage caused by alcohol in mice.Experiment 4: The effect of lyophilized very fine powder of peony petals in the prevention and treatment of chronic alcoholic liver injury in mice.Sixty mice were randomly divided into controls,models,biphenyl diesters,peony petals lyophilized very fine powder low,medium and high dose groups.The control group and the model group were gastricly filled with equal volume distilled water,and the remaining groups were gastrically filled with150 mg/kg biphenyl diester and 80 mg/kg,160 mg/kg,320 mg/kg peony petals lyophilized very fine powder,0.5 hours after each administration,except for the control group,the remaining groups were gastricly filled with red star two pot heads(the first week used 40%of the 56-degree liquor,the concentration of the liquor was increased by 5% in 2-3 weeks,and the 56-degree liquor was started in the fourth week;the dose for the first 2 weeks was10 m L/kg,The dose is 12 m L/kg for the next 2 weeks).1 time per day for 30 days.The results showed that the freeze-dried very fine powder group of peony petals could significantly improve the abnormal conditions of liver,stomach,heart and brain coefficients caused by alcohol in mice,significantly reduce the content of AST,ALT and TG and MDA in serum,increase the activities of ASH and SOD in liver tissue and GSH-PX and CAT in gastric tissue,reduce CK and LDH activity in heart tissue,increase GSH activity and decrease NO content in brain tissue,and increase the liver,stomach,brain,Observation of histopathological sections such as the heart confirmed that compared with the model group,edema and inflammation were significantly alleviated in each group of lyophilized very fine powder of peony petals,especially in the medium dose group,the pathological improvement was the most significant(basically returned to normal level).Conclusion: For the first time,the nutrients in the lyophilized very fine powder of Shaanxi Heyang peony petals were detected,and it was confirmed that the nutrients contained in them were relatively complete,especially the vitamin C and the necessary amino acids of the human body were high,and they were also rich in essential chemical elements.The content of 6 chemical components such as gallic acid in the freeze-dried extremely fine powder of peony petals was clarified.It was confirmed that the lyophilized ultra-fine powder of peony petals had a significant effect on the prevention and treatment of both alcohol-induced acute and chronic liver injury in mice. |
