Objective To explore the characteristics and rules of the blood perfusion of BL13 in rats in response to the pathological changes of chronic obstructive pulmonary disease(COPD),and further explore the effect of electroacupuncture(EA)on the local blood perfusion of BL13 and the pathological progress of COPD.Method Experiment 1 Study on the early response of blood perfusion at BL13 point to the pathological changes of COPD SPF healthy male Wistar rats were randomly divided into 5-day model group(group C1),10-day model group(group C2),15-day model group(group C3),20-day model group(group C4),25-day model group(group C5),30-day model group(group C6)and normal group(group N).COPD was induced in group C by exposure to cigarette smoke combined with airway drip of lipopolysaccharide,and group N was normally fed during the modeling period.Blood perfusion of BL13 and points beside BL13 were monitored by a laser speckle imaging system(PeriCam PSI)before modeling,on the 5th,10th,15th,20th,25th and 30th day of modeling.After modeling,the lung tissue and alveolar lavage fluid of these groups were taken for pathological sections of lung tissue and ELISA detection of IL-1β and IL-6.Finally,we need to analyze the correlation between the blood perfusion of BL13 and the evaluation index of COPD model.Experiment 2 Study on the changes of blood perfusion of BL13 in rats during the whole cycle of COPD SPF healthy male Wistar rats were randomly divided into model group(group C),treatment group(group A)and normal group(group N).COPD was induced in group C and group A by exposure to cigarette smoke combined with airway drip of lipopoly saccharide.After 30 days,group A and C were stopped modeling and group A was given ambroxol hydrochloride oral solution(100ml:0.6g)once a day(5mg/kg)for 3 weeks while group C was given equal volume of saline and group N was fed normally in the whole process.Blood perfusion of monitoring points(BL13,points beside BL13,LR3 and GB34)was recorded before modeling,on the 15th,30th day of modeling,and at the end of three weeks of treatment.The methods of material selection,model evaluation and correlation analysis after modeling are the same as before.Experiment 3 Effect and intervention of electroacupuncture of BL13 on blood perfusion of it in COPD rats SPF healthy male Wistar rats were randomly divided into model group(group PPE),electroacupuncture model group(group PPE+EA),normal group(group N),electroacupuncture normal group(N+EA group).On the first day of modeling,group PPE and PPE+EA were treated with airway drip of porcine pancreatic elastase(PPE)solution,and fed normally as group N and group N+EA in the next month.Group PPE+EA and N+EA received electroacupuncture for two weeks since the second day of modeling.Blood perfusion of monitoring points(BL13 and points beside BL13)was recorded before modeling,on the 15th,18th,21th,24th,27th,30th day of modeling.The methods of material selection,model evaluation and correlation analysis after modeling are the same as before.Result1.Results of experiment 1 showed that:① Blood perfusion of BL13 and lung pathology:After modeling,the blood perfusion of left and right BL13 in group C increased from the 5th day and was always higher than that in group N.On the 10th day,the blood perfusion of BL13 in group C reached or approached the peak value,and remained statistically different from that in group N until the end of modeling(P<0.05,P<0.01).In the process of modeling,the pathological condition of lung in group C was gradually aggravated,and there were differences between group C and group N(P<0.05,P<0.01).Concentration of IL-1β and IL-6 respectively peaked on the 10th and 5th day,corresponding to the changes of BL13 blood perfusion.② Blood perfusion of the point beside BL13:Except that the blood perfusion of the point beside BL13 in group C peaked on the 10th day and significantly higher than group N(P<0.01),there were no difference between them at other modeling time(P>0.05).③Correlation analysis:From the 10th day of modeling,the positive correlation between blood perfusion of BL13 and lung pathology,blood perfusion of the point beside BL13 and lung pathology was gradually obvious and significant(P<0.05,P<0.01,P<0.001),but there were more times of strong correlation between blood perfusion of BL13 and lung pathology.2.Results of experiment 2 showed that:① Blood perfusion of BL13 and lung pathology:During the three periods of occurrence,development and recovery of COPD,blood perfusion of left and right BL13 in group C and group A was always higher than that in group N(P<0.05,P<0.01).After stopping modeling,the blood perfusion of BL13 in group C decreased with the natural recovery of the disease,but the blood perfusion decreased more significantly in group A after treatment intervention.At the end of treatment,the blood perfusion of BL13 in group A was significantly lower than that in group C(P<0.05,P<0.01).The final severity of lung pathology in each group also showed the trend of "group C>group a>group N",and there was significant difference between the groups(P<0.05,P<0.01).② Blood perfusion of the point beside BL13:There was no difference between blood perfusion of the point beside BL13 of those groups except that the blood perfusion of group C and group A was significantly higher than that of group N on the 30th day of modeling(P<0.01).③ Blood perfusion of LR3 and GB34:there was no regular change in blood perfusion of LR3 and GB34 in each group,and there was no difference between the groups(P>0.05).④ Correlation analysis:There were significant positive correlations between blood perfusion of BL13 and lung pathology(P<0.05,P<0.01,P<0.001),while the correlation between blood perfusion of the point beside BL13,GB34 and LR3 blood perfusion and lung pathology was low(P>0.05).3.Results of experiment 3 showed that:① Blood perfusion of BL13 and lung pathology:the blood perfusion of BL13 in group PPE+EA increased briefly after acupuncture,and decreased significantly since stopping acupuncture.The blood perfusion of BL13 in group PPE was always higher than that in other groups at the later stage of modeling and always has significant differences compared with group N and group N+EA(P<0.01,P<0.05),while there were differences between group PPE and group PPE+EA sometimes(P<0.01,P<0.05).The severity of lung pathology at the end of modeling in those groups also showed the trend of"PPE group>PPE+EA group>N group and N+EA group",and there were significant differences between groups(P<0.01,P<0.05),which were consistent with the change trend of blood perfusion of BL13.② Blood perfusion of the point beside BL13:Blood perfusion of the point beside BL13 in group PPE and group PPE+EA was higher than that in group N and group N+EA in the middle and late stage of modeling,and showed some differences(P<0.01,P<0.05),but the regularity was not strong.③Correlation analysis:There were strong positive correlations between blood perfusion of BL13 and lung pathology,also between blood perfusion of the point beside BL13 and lung pathology(P<0.05,P<0.01,P<0.001).Conclusion1.The preliminary results show that the blood perfusion of BL13 has the characteristics and laws of early response and whole process response to the pathological changes of COPD.2.Early electroacupuncture at BL13 can delay the pathological progress of COPD,and does not affect the characteristics of local blood perfusion at BL13 in response to lung pathological changes.
