Objective:In this study,toll like receptor-2(TLR-2)and human α-The expression and significance of defensin-3(HNP-3)in women with premature rupture of membranes(PROM)in Qinghai area,that is,premature rupture of membranes in Qinghai area,provides a certain experimental basis for the research and prevention of premature rupture of membranes,and finds a new breakthrough in the prevention of premature rupture of membranes.Methods:(1)The fetal membrane tissue samples of PROM pregnant women and normal pregnant women who were hospitalized and delivered in the Affiliated Hospital of Qinghai University from June 2020 to June 2021 were collected.There were 30 cases in PROM group and 30 cases in normal pregnancy group.(2)The localization and expression of TLR-2 and HNP-3 in PROM and fetal membranes of normal pregnant women were detected by immunohistochemistry;The expression levels of TLR-2 and HNP-3 in PROM and fetal membranes of normal pregnant women were detected by Western blot.(3)The expression of TLR-2 and HNP-3 in fetal membranes between the two groups were analyzed by independent sample t-test;The correlation between the two variables was tested by human correlation test.Results:(1)Immunohistochemistry showed that TLR-2 and HNP-3 were expressed in PROM group and normal pregnancy group in Qinghai area,and the localization of the two groups was basically the same;The average optical density of TLR-2 and HNP-3 in PROM group was higher than that in normal pregnancy group(P < 0.05);Western blot showed that the protein expression levels of TLR-2 and HNP-3 in PROM group were higher than those in normal pregnancy group(P < 0.05);(2)Western blot showed that the expression levels of TLR-2 and HNP-3 in PROM group were positively correlated(r = 0.400,P = 0.029);There was no correlation between TLR-2 and HNP-3 protein expression in normal pregnancy group.Conclusion:(1)TLR-2 and HNP-3 were expressed in the fetal membrane tissue of the PROM group and the normal pregnancy group in the Qinghai region,and the two groups were basically the same,while the expression level of TLR-2 and HNP-3in the fetal membrane tissue of the PROM group was higher In the normal pregnancy group,it is preliminarily considered that TLR-2 and HNP-3 may be involved in the occurrence of PROM in this region.(2)The expression level of TLR-2 and HNP-3 in the PROM group in the Qinghai area were positively correlated,and there was no significant correlation in the normal pregnancy group.It is considered that the two may be involved in the occurrence of PROM in this area,but the sample size of this experiment is small,further research is needed.
