Objective:To investigate the expression of miR-146b and Her-1 in gastric cancer and their correlation,and to study the relationship between the their expression level and clinicopathological features.Our research may provide some evidence for exploring the occurrence mechanism and molecular targeted therapy of gastric cancer.Methods:A total of 40 patients with gastric cancer first confirmed by gastroscopy and pathological biopsy were collected,and we collected gastric cancer tissues and paired normal tissues.RT-q PCR was used to determine the relative expression of miR-146b and IHC was used to determine the expression of Her-1.Finally,we analyzed the correlation between the expression of Her-1 and miR-146b in gastric cancer,as well as the correlation between the their expression and clinicopathological features respectively.Results:1.The relative expression level of miR-146b in gastric cancer tissues was lower than paired paracancer tissues(P<0.01).The expression level of miR-146b in gastric cancer tissues had statistically significant difference in patients’age,tumor differentiation degree,clinical stage,lymph node metastasis and intestinal metaplasia(P<0.05).There were no significant differences in gender,tumor site,tumor diameter,invasion depth and Hp infection(P>0.05).2.The expression level of Her-1 in gastric cancer tissues was significantly higher than paired paracancer tissues(P<0.001).The expression level of Her-1 in gastric cancer tissues showed statistically significant differences in tumor differentiation,lymph node metastasis,invasion degree and clinical stage(P<0.05).There were no significant differences in gender,tumor site,tumor diameter,Hp infection and intestinal metaplasia(P>0.05).3.Spearman rank correlation analysis showed that there was a negative correlation between the expression of miR-146b and Her-1 in gastric cancer tissues(r_s=-0.348,P<0.05).Conclusions:1.The expression of miR-146b was down-regulated and Her-1was up-regulated in gastric cancer tissues,and their expression level was correlated with some clinicopathological features,suggesting that miR-146b and Her-1 may be involved in the occurrence and development of gastric cancer.2.miR-146b may play a role as a tumor suppressor gene in gastric cancer,inhibiting the occurrence and development of gastric cancer,and Her-1 may promote the occurrence and development of gastric cancer,which may provide certain clinical evidence for the diagnosis and molecular targeted therapy of gastric cancer.3.The expression of miR-146b and Her-1 showed a weak negative correlation in gastric cancer tissues,although the causal relationship between them cannot be determined,it is of certain significance to study the occurrence mechanism and targeted therapy of gastric cancer.
