Expression Of SCO1 And SCO2 After Form-deprivation Myopia In Guinea Pigs | | Posted on:2023-10-05 | Degree:Master | Type:Thesis | | Country:China | Candidate:G M Zhou | Full Text:PDF | | GTID:2544306911477534 | Subject:Medical Technology | | Abstract/Summary: | PDF Full Text Request | | Objectives:To observe the morphologic changes in the retina and sclera of guinea pigs with form-deprivation myopia(FDM)and examine the expression of SCO1 and SCO2 in the retina and sclera of guinea pigs with FDM.To explore the relationship between SCO1 and SCO2 genes and FDM.Methods:The 3-week-old tricolour guinea pigs were selected as the experimental sample,which was randomly divided into a blank control group(n=18)and an experimental group(n=18).The right eyes of the experimental group were used as the FDM group and the left eyes were used as the self-control group,while the guinea pigs in the blank control group were left untreated.The refraction and axial length of the guinea pigs were measured at weeks 0,1,2,3 and 4 after modeling.After 4 weeks,the guinea pigs were executed.The morphologic changes of the retina and sclera were observed by HE staining;the relative expression of collagen fibers in the sclera of each group was measured by Masson staining;the relative expression of SCO1 and SCO2 proteins in the retina and sclera of each group was measured by Western blot and immunohistochemistry;the relative expressions of SCO1 and SC02 mRNA in the retina and sclera of guinea pigs were measured by RT-qPCR.Results:After 4 weeks of form deprivation,the refraction in the FDM group changed from(4.184±0.725)D to(-1.796±0.466)D.The degree of myopia was significantly higher than that in the self-control group(2.649±0.566)D(P<0.001)and the blank control group(2.322±0.802)D(P<0.001);the axial length(8.520±0.189)mm was significantly longer than that in the self-control group(7.871±0.353)mm(P<0.001)and the blank control group(7.832±0.225)mm(P<0.001).The retinal and scleral tissues were moderately thinner,and the ganglion cells and the cells of inner and outer nuclear layers in the retina became fewer.The collagen content of the sclera became less in the FDM group compared with the self-control group(P<0.001)and blank control group(P<0.001).The relative expression of SCO1 and SCO2 proteins and the SCO1 and SCO2 mRNA in the sclera and retina was significantly lower in the FDM groups than in the self-control group and blank control group(all P<0.05);there was no statistical significance in the relative expression of SCO1 and SCO2 proteins and SCO1 and SCO2 mRNA between the self-control group and blank control group(all P>0.05).Conclusions:After 4 weeks of form deprivation,the axial length of the guinea pig elongated and the refraction drifted significantly towards myopia;the retina and sclera became thinning,and the retinal and scleral morphology became changing.The relative expression of SCO1 and SCO2 proteins and SCO1 and SCO2 mRNA in the retina and sclera of guinea pigs with deprivation myopia decreased,and the SCO1 and SCO2 genes were involved in the development of deprivation myopia through some mechanism. | | Keywords/Search Tags: | form-deprivation myopia, synthesis of cytochrome c oxidase 1, synthesis of cytochrome c oxidase 2, guinea pig | PDF Full Text Request | Related items |
