| Objective:Crohn’s disease,one kind of inflammatory bowel disease,affects millions of patients all over the world.The research on Crohn’s disease(CD)is not sufficient in the field of traditional Chinese medicine(TCM).There are few reports on large sample study of TCM syndrome type.We used bibliometrics visualization analysis in order to understand the research hotspot of CD in the field of TCM during the past 20 years.We analyzed clinical data to explore the distribution of syndrome types of CD in TCM and its relationship with oxidative stress index(SOD and MDA).The core genes related to oxidative stress in CD were screened by biogenic analysis.We explored the possible mechanism and the potential treatment of TCM by analyzing these genes.Methods and Results:Articles related to TCM research in CD were used for bibliometric analysis,which downloaded from CNKI,Wanfang,VIP and Web of Science(WOS)by CiteSpace software.We found that main key words of the Chinese articles were theories of TCM,treatment measures,mechanism exploration and curative effect comparison.English literature focused on mechanism exploration and experimental studies.The distribution characteristics of TCM syndromes was analyzed from the clinical data of 210 patients with CD.TCM syndrome types include liver depression/spleen deficiency(26.67%),Yang deficiency in spleen and kidney(23.33%),dampness/heat connotation(21.43%),dampness/cold trap the spleen(17.14%)and Qi stagnation/lood stasis(11.43%).To explore the correlation between syndrome type and oxidative stress,44 untreated patients and 15 healthy subjects were selected for oxidative stress analysis.We found that the difference of SOD and MDA values between normal group and different TCM syndrome groups were statistically significant(P<0.05).Ten core genes of oxidative stress in CD screened from GEO database were IL1B,ICAM1,PTGS2,CXCL1,CXCL8,TIMP1,MMP3,MMP1,SELE and SELP.28 Chinese herbs were predicted with the function of modulating these genes.In the four-gas analysis,the cold character was 29.63%,the slight cold and warm were 22.22%respectively.The bitterness in five taste analysis was 51.85%.Liver(48.15%),lung(44.44%),spleen(37.04%),and stomach(33.33%)were the main subjects of meridian conversion.The efficacy included clearing heat and detoxifying(18.52%),clearing heat and dampness,promoting bile and eliminating yellow,dispelling wind and dampness,clearing heat and cooling blood(7.41%each).Conclusion:It is valuable to study oxidative stress of CD in the field of traditional Chinese medicine.The oxidative stress of CD is a multi-target and multi-pathway regulated process.We speculated that CD was related to the dampness-heat syndrome type of oxidative stress from medicinal properties.This study indicated that many chemical components in traditional Chinese medicine could treat CD by regulating multi-target and multi-signaling pathways,and had a broad prospect. |
