Objective:Successfully establish rat model of type 2 diabetes and Alzheimer’s disease.Compare the blood glucose, body weight, oxidative stress markers and pathological sections of brain tissue between all treatment groups and Composite model group.To explore the relationship between T2DM and AD pathogenesis,the treatment effect and target of Bushen Jianpi Decoction on T2DM and AD by different diseases with the same therapy.Methods:80 SPF SD male rats, aged 6 weeks, weighing 200+20g, adaptive feeding a week before the experiment began. Divide into six groups:blank group, compound model group, sham operation group, high dose of Chinese medicine group, medium dose of Chinese medicine group, low dose of Chinese medicine group.The compound model group was further made AD mode on the basis of the successful model of T2DM. Copy T2DM model by adopting the method of high-fat diet+intraperitoneal injection of STZ.Copy AD model by injecting A beta 1-42 in bilateral hippocampal CA1 area. The sham operation group was based on the T2DM model, and the bilateral hippocampal CA1 region was injected with the volume of ammonia water.Large, medium and small dose group respectively in the composite model based on given Bushen Jianpi formula by 24,12g,6g/kg/d gavage administration. After 6 weeks of treatment, all rats were sacrificed in serum and brain tissue were measured in serum, hippocampus, cerebral cortex, the SOD activity in the cerebellar cortex, GSH-Px and the content of MDA, and brain tissue pathological slices do he and Congo red staining. In the course of the experiment, weekly measurements of body weight of rats, the blood glucose.Results:1.Successfully establish T2DM and AD compound model.2.Blood glucose:Traditional Chinese medicine large, medium and small dose group,6 weeks after treatment of blood glucose than compound model group has decreased, but no significant statistical difference (P>0.05).3.Weight:Compared with the blank group, the compound model group rats appeared more drink, more urine, weight loss.During the same period, there was a significant difference between the two groups (P< 0.05).Compared with the compound model group, the effect of weight loss in the large, medium and small dose group was improved.The improvement showed dose dependence.4.Indicators of oxidative stress among groups:Compared with the blank group, the SOD and GSH-PX activity in serum, hippocampus, cerebral cortex and cerebellar cortex of the compound model group decreased(P<0.01),and MDA content increased(P<0.01).Compared with sham operation group, the SOD and GSH-PX activity in serum, hippocampus, cerebral cortex and cerebellar cortex of the compound model group decreased(P<0.05),and MDA content increased(P<0.05).Compared with the compound model group, the SOD, GSH-Px activity in serum, hippocampus, cerebral cortex, cerebellum cortex of the large, medium and small dose of Chinese medicine group increased (P< 0.05 or P< 0.01), MDA content decreased (P< 0.05 or P< 0.01).The improvement showed dose dependence.5.The different parts of oxidative stress in brain tissue of the same group rats:In the same group of rats, the difference of SOD activity in hippocampus, cerebral cortex and cerebellar cortex was not statistically significant (P>0.05).In the same group of rats, the difference of MDA content in hippocampus, cerebral cortex and cerebellar cortex was not statistically significant (P>0.05).In the same group of rats, the difference of GSH-Px activity in hippocampus, cerebral cortex and cerebellar cortex was statistically significant(P<0.01).6.Pathological section:HE staining showed that a large number of neurons in the hippocampus of the model group were changed, the arrangement of the cells was disordered, the neurons were fixed and stained, and the apoptotic bodies were found.Sham operation group only cell structure disorder, the basic form of normal neurons.The morphology and arrangement of neurons in the large and medium dose group were improved compared with that in the compound model group. And the improvement of traditional Chinese medicine large dose group is more obvious, the improvement of small dose group of Chinese medicine is not obvious. Results:1.high-fat diet+intraperitoneal injection of STZ can successfully replicate the T2DM model.On this basis, bilateral hippocampal CA1 region injection of A beta 1-42,we can successfully replicate the T2DM combined AD model.2.Bushen Jianpi Decoction, by strengthening the spleen and kidney,rlt can improve the body weight, brain tissue pathology and oxidative stress in type 2 diabetic patients and Alzheimer’s disease.Oxidative stress is one of the common targets of Bushen Jianpi Decoction in the treatment of two diseases.
