Celastrus L.plants is widely distributed in China,except Xinjiang and Qinghai,and it is distributed all over the country,especially in the south of the Yangtze River.The preliminary work of this project found that the ethyl acetate extract(COE)of celastrus orbiculatus has significant anti-digestive tract tumor activity.In this study,different extraction processes of celastrus orbiculatus were investigated by chemical-biological activity analysis.The crude extract of HepG2 human hepatoma cell line with strong in vitro tumor cytotoxic activity was obtained by scale extraction with better extraction technology.Vacuum normal phase and reversed phase silica gel column chromatography and high performance liquid chromatography were used for separation and purification.The structures of 18 monomer compounds were determined by four spectra(NMR,UV,IR,MS),optical rotation,calculation of ECD and comparison of literature.Respectively are Celastorbiol A(1),Celastorbiol B(2),Celastorbiol C(3),7,4’-dihydrohomoisoflavanone(4),3-deoxysappanone B(5),sappanone A(6),3’-deoxysappanone A(7),bonducelin(8),isoliquiritigenin(9),4’-Omethyldavidigenin(10),liquiritigenin(11),(-)-butin(12),apigenin(13),luteolin(14),3’,4’,7-trihydroxyflavone(15),acacetin(16),diosmetin(17),1,3-propanediol,2-amino 1-(4-hydroxy-3-methoxyphenyl)(18).Among them,1-3 are new compounds,2-17 are flavonoids,and 1 and 18 are phenylpropanoid compounds.In the later stage,more homologues will be isolated and identified.Together with the monomer compounds that have been isolated and identified so far,the antitumor activity will be studied.In this study,18 secondary metabolites have been isolated and identified from celastrus orbiculatus,which will provide material basis for the study of anti-tumor efficacy and pharmacological mechanism of subsequent monomer compounds,as well as the study of anti-tumor activity after the combination of monomer compounds.
