Objective: Platelet rich fibrin(PRF)was prepared from autologous blood of the New Zealand rabbits.The rib and auricular cartilage of rabbits was wrapped with PRF(Platelet-rich Fibrin)membranes and grafted into the back fascia of the rabbits.To explore the effect of PRF on the transplantation of New Zealand rabbit rib and auricular cartilage.Methods: Fifteen rabbits were selected.After satisfactory anesthesia,the venous blood of rabbits was taken and the blood was immediately centrifuged at 3000 rpm for 10 min to make PRF.PRF squeezed excess water was prepared for PRF membrane.Two sets of costal cartilage and auricular cartilage of each rabbit were obtained by surgery,and the pericardium of cartilage was removed.Take costal cartilage and auricular cartilage wrapped with PRF membrane implanted into the dorsal facia of rabbits as costal cartilage and auricular cartilage experimental group.Another group of costal cartilage and auricular cartilage were directly implanted into fascia layer as the costal cartilage and auricular cartilage control group.Each five rabbits were sacrificed at 4th,8th and 12 th week.The all cartilage of the four groups was photographed,weighed,stained by safranin O-solid green,alcian blue,hematoxylin-eosin,and immunohistochemistry of type II collagen.The weight of cartilage,the number of chondrocytes,the area of chondrocytes,the number of blood vessels aroundthe cartilage and the positive proportion of type Ⅱ collagen staining were statistically analyzed.Results: The surface of the costal cartilage and auricular cartilage experimental group was wrapped by dense smooth fibrous tissue which is thicker than that of control group.As the implantation time went on,the weight,chondrocytes area and number of costal cartilage and auricular cartilage decreased than before experiment,and there was no significant difference between the costal cartilage and auricular cartilage experimental group and the costal cartilage and auricular cartilage control group,indicating that PRF had no significant effect on the weight,cell number and area of cartilage.Under microscope,the cartilage surface of the experimental group was thickened,which was fibrous tissue,and some costal cartilage and auricular cartilage experimental groups had angiogenesis around the cartilage.The costal cartilage and auricular cartilage experimental group produced more type 2 collagen than the costal cartilage and auricular cartilage control group,indicating that PRF could promote the secretion of type 2 collagen by chondrocyte.Conclusion: In the autologous cartilage heterotopic transplantation,PRF could promote the production of type II collagen by chondrocytes.The combination of PRF and cartilage formimg the capsule around the cartilage could increase the volume of graft.The PRF could promote the angiogenesis around cartilage.Within 12 weeks,the quality of transplanted costal and auricular cartilage decreased gradually with time.PRF had no significant effect on cartilage quality,cell size,number and morphology.PRF had no significanteffect on the survival of cartilage at early stage.
