Objective:This experiment was conducted to evaluate the superior frequency of acupuncture for IBS by observing the effects of different frequencies of electroacupuncture bilateral Zusanli(ST36)on colonic sensitivity, visceral nociceptive threshold, colonic slow wave discharge and other colonic functions as well as serum 5-HT,Ach and anxiety status in rats, and to provide a reference basis for optimizing and standardizing the clinical protocol of acupuncture for IBS.Methods:100 SPF-grade Wistar rats weighing 200±10 g were fed for one week after acclimatization,15 rats were randomly selected as the blank group using the random number table method, and the remaining rats were prepared as IBS models using the 10-day chronic water avoidance stress method, and the success of the model preparation was judged using the semi-quantitative scoring method of the abdominal wall retraction reflex of the colorectal dilatation assay, and the successful modeling rats were subsequently randomly The rats were then randomly divided into model group, treatment group A and treatment group B,with 15 rats in each group using the randomized number table method.Treatment A group was treated once a day, while treatment B group was treated once every two days, and both groups were treated 10 times. The AWR scores, behavioral indices of the elevated cross maze,as well as the visceral nociceptive thresholds, proximal colonic slow wave electrical rhythm and serum 5-HT and Ach levels of rats in each group after treatment were collected and analyzed before modeling, after modeling and after treatment.Results:(1)After treatment, the visceral pain sensitivity of rats in both treatment groups was significantly reduced(P<0.05),and the visceral nociceptive threshold was significantly improved(P<0.05);(2)After treatment, the colonic slow wave rhythm of rats in both treatment groups was well restored, and the frequency and amplitude of colonic slow waves were significantly decreased compared with the model group(P<0.05);(3)After treatment, rats in both treatment groups had their The behavioral performance of the rats in both treatment groups showed that their symptoms of anxiety were improved(OE:P<0.05);(4)The results of serum 5-HT and Ach levels showed that both treatment groups were able to reduce the serum 5-HT and Ach levels in IBS rats;(5)There was no significant difference between the two treatment groups in the comparison of the above test indicators.Conclusion:(1)The frequency of acupuncture once a day and twice a day can improve the visceral pain sensitivity, colonic slow wave electrical rhythm and serum 5-HT,Ach and other indicators of IBS rats.(2)There was no significant difference between the two acupuncture frequencies in improving the colon function of IBS rats.(3)The preliminary results of this experiment suggest that both doctors and patients can avoid the psychological burden of worrying about the disease when they can not be treated in time due to the delay of events.Doctors and patients can choose the treatment frequency flexibly, but ensure that the treatment frequency is sufficient.
