| Objective: Based on Tamm-horsfall protein(THP)to study the effect and mechanism of the treatmentthe of mongolian medicine Huo Xiao Qi Wei powder of mice on calcium oxalate kidney stones caused by ethylene glycol and ammonium chloride can provide scientific basis for its clinical application.Methods: 72 SPF KM male mice were selected,and the eligible mice were randomized into6 groups according to their basal body weight: normal group,calcium oxalate kidney stones model group,potassium sodium hydrogen citrate granules positive control group,mongolian medicine Huo Xiao Qi Wei powder high-,medium-,and low-dose groups,and 12 mice in each group.The normal group drank water freely /d,model group,positive control group and mongolian medicine Huo Xiao Qi Wei powder high-,medium-,and low-dose groups received 1m L/d 1% ethylene glycol + 1% ammonium chloride by gavage and continuously for 4 weeks to replicate the calcium oxalate kidney stone model.From the 5th week,mongolian medicine Huo Xiao Qi Wei powder high-,medium-,and low-dose groups were given Huo Xiao Qi Wei powder1.0g/kg,0.5g/kg,0.25g/kg /d and the positive control group received 1.67 g / kg /d potassium sodium hydrogen citrate granules by gavage,and the other groups drank water freely/d.Results:(1)The results of the effect of mongolian medicine Huo Xiao Qi Wei powder on the urine volume of mice showed that compared with the model group,the urine volume increased with the statistically significant difference(P <0.05).(2)The results of the effect of mongolian medicine Huo Xiao Qi Wei powder on kidney histopathology of mice showed that the HE staining scores of kidney histopathological sections of each administration groups of mice was less severe than the model group.Mongolian medicine Huo Xiao Qi Wei powder groups were compared with the model group,the pathology score was significantly reduced(p <0.05).(3)The results of the effect of mongolian medicine Huo Xiao Qi Wei powder on the kidney coefficient in mice showed that the mongolian medicine Huo Xiao Qi Wei powder groups were compared with the model group,the kidney coefficient became smaller and the difference was statistically significant(p <0.05).(4)The results of the effect of mongolian medicine Huo Xiao Qi Wei powder on THP in mouse urine showed that the mongolian medicine Huo Xiao Qi Wei powder groups were compared with the model group,urinary THP had the effect of restoring its expression and the difference was statistically significant(p <0.05).Conclusion: Mongolian medicine Huo Xiao Qi Wei powder has a therapeutic effect on mouse of kidney stone model,and its mechanism is related to the recovery and expression of THP in urine,thus inhibiting the aggregation,growth and nucleation of urine crystallization. |
