| BackgroundLung cancer has always been a worldwide health problem,and its prevalence and mortality are extremely high in all cancer types.ferroptosis is a unique form of programmed cell death,and there is a large body of literature indicating that ferroptosis plays an important role in a variety of diseases in multiple systems,while SLC7A11(solute carrier family 7 member 11)is a key protein regulating ferroptosis inhibitory pathways.The diagnostic and prognostic value of SLC7A11 and its association with immune cell infiltration in lung cancer remains unclear.In this study,we found that SLC7A11 was overexpressed in lung cancer tissues using a variety of public open database analysis.There was no significant difference in the expression of SLC7A11 between NSCLC patients with different TNM stages,smoking history and P53 mutation.SLC7A11 expression is associated with the prognosis of lung cancer patients.Using ROC curve analysis,SLC7A11 may be a potential tumor diagnostic marker.Furthermore,SLC7A11 expression was inversely correlated with the level of infiltration of immune cells and the expression of multiple immune cell markers.Importantly,SLC7A11 may affect the prognosis of LUAD patients in part due to immune infiltration.In conclusion,SLC7A11 may play a significant role in the progression of lung cancer and the regulation of immune cell infiltration.ObjectiveThe aim of this study was to analyze the expression of SLC7A11 in non-small cell lung cancer(NSCLC)using a variety of public open databases,to analyze the relationship between the expression of SLC7A11 and clinical features,and the diagnostic and prognostic value of SLC7A11 in lung cancer.In this study,we also analyzed the relationship between SLC7A11 expression and patient prognosis.Secondly,the relationship between SLC7A11 expression and immune infiltration in lung cancer was analyzed to further explore whether the poor prognosis of LUAD patients was caused by immune infiltration.By summarizing the literature research results,it is proved that SLC7A11 plays a role in the diagnosis and prognosis of lung cancer,and SLC7A11 can specifically affect the immune infiltration of lung cancer.MethodThe relationship between SLC7A11 expression and tumor type,tumor stage,and patient characteristics was analyzed using online public databases such as TCGA,TIMER,GEPIA2,UALCAN,Kaplan-Meier Plotter,and TISIDB.Kaplan-Meier curves were used to analyze the relationship between SLC7A11 expression and patient prognosis.ROC curves were used to assess the role of SLC7A11 in the prediction of lung cancer diagnosis.Protein-protein interaction network(PPI),genegene interaction network and significantly differentially expressed genes were analyzed using STRINGE and Genemania databases,and the top 50 genes similar to SLC7A11 in lung cancer were obtained in the GEPIA2 public database,and these genes were enriched and analyzed by Gene Ontology(GO)and KEGG(Kyoto Encyclopedia of Genes and Genomes)using the "ClusterProfiler" software package,and the corresponding genes were visualized by the "ggplot2" software package.Results from Kaplan-Meier curves were analyzed using the log-rank test.Correlation of gene expression was evaluated by Spearman correlation and statistical significance.P-values<0.05 were considered statistically significant.Result1.The expression of SLC7A11 increased in lung adenocarcinoma and squamous cell carcinoma(P<0.05).There was no significant difference in the expression of SLC7A11 among different clinical stages,lymph node metastasis,smoking history,and P53 mutations.2.K-M curve analysis showed that a poor overall survival(OS)and progression free survival(PFS)in NSCLC patients with high expression of SLC7A11.3.The differential expression level of SLC7A11 may be a prospective biomarker for diagnosis.4.Analysis of similar genes in SLC7A11 by GO and KEGG showed that SLC7A11 is related to the response to oxidative stress,toxic substances,and cell detoxification.These similar genes are mainly involved in ferroptosis pathways.5.The poor prognosis of lung adenocarcinoma patients with high expression of SLC7A11 may be caused by affecting partial immune infiltration.Conclusion1.The expression of SLC7A11 is increased in NSCLC patients,and the expression of SLC7A11 in male is higher than that in female.2.SLC7A11 may be a potential diagnostic and prognostic biomarker for NSCLC;3.SLC7A11 may affect immune infiltration by participating in ferroptosis pathways in NSCLC. |
