The Role Of P2RX1 In The Prognosis And The Mechanism Of Immune Infiltration In Patients With Serous Ovarian Carcinoma | | Posted on:2024-08-12 | Degree:Master | Type:Thesis | | Country:China | Candidate:Z M Song | Full Text:PDF | | GTID:2544306929974709 | Subject:Clinical Laboratory Science | | Abstract/Summary: | PDF Full Text Request | | ObjectiveThis study aimed to investigate the relations between P2RX1 expression and clinicopathological factors and tumor immune infiltration in OV.This may provide new clues for prognosis and immunotherapy of OV patients.Methods(1)The expression of P2RX1 in ovarian cancer tissue was detected by IHC;the m RNA expression of P2RX1 was investigated by q RT-PCR technology;the expression of P2RX1 in serum samples of ovarian cancer was detected by ELISA.(2)Download and analyzed the RNA transcript data of P2RX1 in TCGA database,combined with P2RX1 expression data such as CCLE and CPTAC,and comprehensively analyzed the expression pattern of P2RX1 in OV.The R software was used to drawing different plots.Integrated the expression profile data of P2RX1 in TCGA and clinical information of patients,and analyzed the correlation between its expression and different clinicopathological factors of OV patients.The expression profile data were divided into high and low quartile expression groups,and the differentially expressed genes were screened in order to explore the signal pathways and biological functions.The “x Cell” and“EPIC” algorithm were used to analyze the correlation between P2RX1 expression in OV and different infiltrating immune cells and matrix components.The correlation between P2RX1 and immune checkpoints and 60 immune regulatory factors was analyzed by Spearman correlation test.TIDE algorithm was used to analyze the correlation between P2RX1 and immune checkpoint blockade(ICB)and tumor mutation burden(TMB).Results(1)Immunohistochemical results showed that the protein level of P2RX1 was down-regulated in OV.QRT-PCR results showed that the m RNA expression of P2RX1 was also decreased in OV.ELISA results showed that the expression of P2RX1 was down-regulated in the serum of ovarian cancer patients.(2)The TCGA database showed that the m RNA level of P2RX1 was significantly down-regulated in OV.The CCLE database showed that the expression level of P2RX1 in OV90 was the highest in 45 ovarian cancer cell lines.Patients with higher P2RX1 expression had longer overall survival,disease-specific survival,and survival between disease-free and progression free periods.P2RX1 was negatively correlated with tumor purity;was correlated with immune regulatory and tumor metabolism signaling pathways(P<0.05);was positively correlated with the degree of infiltration of various immune cells,especially B cells was positively correlated with immune score and stromal score;was associated with a variety of chemokines,chemokine receptors,immunosuppressive molecules,immunostimulatory molecules,MHC genes,and multiple immune checkpoints.The higher the expression of P2RX1,the higher the TIDE score of OV patients,that is,the worse the effect of ICB treatment.P2RX1 was positively correlated with TMB in OV patients.ConclusionsP2RX1 might be involved in tumor infiltration in OV patients,and it may be provide new therapy ideas for OV patients. | | Keywords/Search Tags: | P2RX1, ovarian serous carcinoma, prognosis, immunohistochemistry, immune infiltration, immunotherapy | PDF Full Text Request | Related items |
