| Objective: Hyperthyroidism is characterized by drying heat of Yin deficiency.According to statistics,syndrome types related to Yin deficiency account for about40.5% in hyperthyroidism.Therefore,based on the literature data of Yin deficiency hyperthyroidism,this study analyzed the data of Chinese medicine treating Yin deficiency hyperthyroidism with the help of data mining technology,so as to find the medication rules and core drugs of Chinese medicine treating this disease.Network pharmacology of core drugs was conducted to analyze their effective components,effective targets and corresponding pathways in the treatment of hyperthyroidism.Methods:Through searching the clinical research literature on the treatment of Yin deficiency type hyperthyroidism by TCM and integrated Chinese and western medicine published in the past 20 years from China National Knowledge Network(CNKI),VIP Chinese Journal Database and Wanfang database,qualified literature data were selected through inclusion and exclusion criteria.After standardized data collation,the whole into the ancient and modern medical records cloud platform website.Frequency statistics,association analysis,cluster analysis and complex network analysis were conducted on the above data to summarize the medication rules and characteristics of traditional Chinese medicine for Yin deficiency type thyroidism,providing reference for clinical treatment of Yin deficiency type hyperthyroidism.Network pharmacology study of core drugs.Firstly,the active ingredients and targets of the drug were found in the TCMSP database,the chemical professional database and the source network,and the literature was searched.For the active ingredients of some missing targets,the Swiss Target Prediction database was used to predict their targets,and all targets were converted to the corresponding gene abbreviation through the Uni Prot database.The three disease databases of Gene Cards,Drugbank and OMIM were used to search for disease targets,and the common target intersection diagram of core drugs and diseases was constructed.Then,VENNY 2.1 website was used to draw the ’ common target Wayne map ’,and Cytoscape software was used to construct the network diagram of ’ core drug-active ingredient-target ’ and ’hyperthyroidism-related component-target ’,and to find the core Chinese medicine components of core drugs for the treatment of hyperthyroidism.Then,the STRING platform was used to construct the PPI network of the common targets,and the screening conditions were set by Cytoscape to find the key targets of the core prescription for the treatment of hyperthyroidism.Finally,the common targets were analyzed by DAVID website to obtain GO and KEGG analysis results.Results: According to the inclusion and exclusion criteria,the literature on the treatment of Yin deficiency type hyperthyroidism was screened,and 128 literatures were included,including 141 prescriptions.The literature data of Yin deficiency hyperthyroidism were analyzed.1.Frequency analysis: TCM syndrome distribution: 40.42% heart-liver Yin deficiency syndrome,27.66% Qi-yin deficiency syndrome,21.99% Yin deficiency Yang hyperactivity syndrome,9.93% liver stagnation Yin deficiency syndrome.The common TCM treatment methods were nourishing Yin and reducing fire 67 times(47.52%),nourishing qi and nourishing Yin 45 times(31.91%),nourishing Yin and purging Yang 19 times(13.48%),eliminating phlegm and dispersing complications 15 times(10.64%).Common drugs include:raw rehmannia Radix,ophiopogon ophiopogon,white peony root,prunella prunus,astragalus,scrophulariae,oyster,schisandra schisandra,Thunberg Fritillaria,cooked Rehmannia,etc.The four drugs are mainly cold,slightly cold,flat,warm;Five common sweet,bitter,Xin taste;Chinese medicine mainly enters the liver,lung,kidney and heart channels.2.Association rules: Radix Astragalus-Radix Rehmanniae,Radix scallops-Radix ophiopogonis,Radix Schisandra-Radix ophiopogon,Radix angelicae-Radix Rehmanniae,Radix coptidis-Radix Rehmanniae,Fritillaria thunbergiae-Radix Rehmanniae,Radix Rhizae thunbergiae-Radix Rehmanniae,Radix Angelicae-Radix Rehmanniae cooked,bionychia-Radix Rehmanniae,Radix scutellariae-Radix Rehmanniae,Radix Suanzaoren-Radix ophiopogonis are the main incompatibility among Chinese herbs.According to the association rules of medicine and syndrome,the syndrome of heart-liver-yin deficiency has a high correlation with Radix Rehmanniae;the syndrome of Qi and Yin deficiency has a high correlation with Radix Astragalus,Radix Rehmanniae and Radix ophiopogonis;the syndrome of Yin deficiency and hyperactivity of Yang has a high correlation with Radix Paeoniae and Radix Xia;the syndrome of liver stagnation and Yin deficiency has a high correlation with Radix Paeoniae and Radix bupleurum,Radix Xia and Radix ophiopogon,poria cocos and Radix scrophulariae.3.Cluster analysis: Through high frequency drug analysis,we can obtain Chinese ginseng,licorice and wolfberry.Sweet mother,peony bark,yam,dogwood;Keel,Oyster,turtle shell,Raw Rehmannia,Astragalus,Cooked Rehmannia,Angelica sinensis,Scutellaria scutellaria,Phellodendron phellodendron,Coptidis;Radix pseudostellariae,Gardenia,Paeoniae alba,Prunella prunus,scrophulariae,Fritillaria thunberg;Codonopsis,bupleurum,sundried licorice,sour jujube kernel,poria cocos,ophiopogon ophiopogon and schisandra chinensis were commonly used in five groups.4.Complex network analysis of traditional Chinese medicine: The core prescription of traditional Chinese medicine for Yin deficiency hyperthyroidism was extracted,consisting of six traditional Chinese medicines: Radix Rehmanniae,Radix paeoniae alba,Radix scrophulariae,Radix astragalus,Radix ophiopogonis ophiopogon and Prunella axella.5.There are 53 effective components in the core drug,297 targets corresponding to the effective components,and 687 targets corresponding to the disease of hyperthyroidism.They intersect with the effective components of the core prescription,and 67 common targets are obtained,including 55 of Astragalus,36 of Paeoniae Radix Alba,55 of Prunella vulgaris,8 of Rehmanniae Radix,20 of Scrophulariae Radix and 11 of Ophiopogon japonicus.There are 34 effective components of hyperthyroidism in the core drugs,among which quercetin,kaempferol,stigmasterol and β-sitosterol are most closely related to the treatment of hyperthyroidism.There are 15 core targets,including STAT3,TP53,EGFR,PTEN,SRC,IL6,TNF,AKT1,VEGFA,F2,MAPK1,CAT,IL1 B,PPARG,NOS3.After enrichment analysis,148 KEGG pathway results,45 CC results,78 MF results and471 BP results were obtained.Conclusion:1.Modern doctors believe that Yin deficiency type hyperthyroidism is based on Yin deficiency,fire exuberance,qi deficiency,liver depression,and Yang hyperthyroidism;The treatment is based on tonifying Yin,clearing heat,benefiting qi,soothing liver,promoting Yang and symptomatic treatment.The main properties of traditional Chinese medicine are cold,slightly cold and smooth,and the taste of traditional Chinese medicine is sweet,bitter and pungent,which mainly affect the liver,lung,kidney and heart meridians.It is further proved that Yin deficiency hyperthyroidism can be treated from multiple zangfu organs.2.In the related compatibility of traditional Chinese medicine,the effect is mainly to nourishing Yin,supplemented by clearing heat,promoting body fluid and dispersing accumulation.Attention should be paid to treating the disease and seeking the root cause,nourishing Yin and promoting body fluid to strengthen the deficiency of the root cause,clearing the heat and cooling the blood,clearing the heat and drying dampness to remove the pathogen of the standard excess.3.Raw rehmanniae radix,Radix paeoniae,Radix scrophulariae radix,Astragalus membranaceus,Radix ophiopogonis radix and Prunella vulgaris are the core prescriptions for the treatment of Yin deficiency hyperthyroidism.The core medicine also takes nourishing Yin and clearing heat as the treatment method,and also tonifying qi,promoting body fluid,softening the liver and dispersing constipation.4.The core prescription may act on key targets such as STAT3,EGFR,TNF,and IL6 through effective active ingredients such as quercetin,kaempferol,stigmasterol,and β-sitosterol.The regulation of TNF signaling pathway,Fox O signaling pathway,CLRs signaling pathway plays a role in the treatment of hyperthyroidism by improving thyroid hormone metabolism,anti-inflammatory,anti-oxidation,immune regulation,anti-cancer and other multiple ways. |
