| Objective:To observe the effects of pulsed electromagnetic field on cartilage of knee joint inflammation of rat,and explore the effect of pulsed electromagnetic fields on cartilage autophagy,serum inflammatory response and m TOR signaling pathway.Methods:1.24 SD male rats were randomly divided into three groups:Control group,osteoarthritis group(OA)and osteoarthritis + pulsed electromagnetic field treatment group(PEMF group),and each group has 8 rats.The OA group and the PEMF group were established by anterior cruciate ligament transportation to mimic traumatic knee arthritis model;2.8 weeks after ACLT surgery,the rats in PEMF group will accept pulse electromagnetic field intervention,with frequency at 8 Hz,intensity at 3.82 mt for 40 minutes/day,5 days/week,8 weeks.The rest rats were put in the pulse electromagnetic field coil without electricity;3.After 8 weeks continuous intervention,all the rats were sacrificed,and the serum was used to detect inflammatory factors via ELISA,the right femurs cartilage were extracted to detect m TOR,Beclin1,MMP13,Adamts5,Aggrecan,Col2a1,SOX9,LC3 B gene expression via PCR,and right tibias were taken to detect Adamts5,Sox9,Col2a1,Beclin1,P62 protein expression via IHC,and left femurs were taken for HE,Safranin-o and fast green staining and OARSI score.Results:1.ELISA analysis: ELISA showed that compared with the Control group,the expression of IL-1β in the OA group was increased,and the difference was statistically significant(P<0.01);compared with the Control group,the expression of IL-10 in the OA group was decreased,and the difference was statistically significant Significance(P < 0.01).2.PCR Analysis: Compared with Control group,ADAMTS5 and MMP13 in OA group was up-regulated,and the differences were statistically significant(P<0.01,P<0.05).Compared with OA group,the ADAMTS5 expression of the PEMF group was down-regulated,and the difference was statistically significant(P<0.05);Aggrecan in OA and PEMF group dropped compared with the Control group,and the differences were statistically significant(P<0.01,P<0.05);COL2A1 in the control group was up-regulated compared to the OA group,and was statistically significant(P<0.01);Compared with the control group,the expression of Beclin1 was increased in the OA group,and the difference was statistically significant(P<0.01).Compared with the Control group,the expression of m TOR in the OA group was decreased,and it was statistically significant(P<0.05).3.Pathological staining and score: Under the optical microscope,most of the stalks of the Control group were complete,and there was partial cartilage shallow.Cartilage stained incompletely and locally with excessive medium cartilage dyeing,and chondrocytes arranged disorderly.Cartilage in OA group destroyed to the middle layer,the tide line is incomplete.There was partially visible new collagen formation,cartilage cell death,and the middle cartilage region has a bone formation.Femur cartilage of PEMF group destroyed less than the OA group,there is no obvious osteoporosis,the cartilage surface is destroyed,and the edge of the cartilage is basically complete,and the stain is clear.Chondrocytes have proliferation without disorders.The OASI score of the OA group increased significantly compared with the Control group,and the difference was statistically significant(P <0.05),while the PEMF group OARSI score was significantly decreased,and the difference was significantly different from the Control group,which was statistically significant(P<0.05).4.IHC Slice: Compared with the Control group,the expression of ADAMTS5 in the OA group was increased,and the difference was not statistically significant(P>0.05);Compared with the OA group,the ADAMTS5 expression in the PEMF group was decreased,and the difference was statistically significant(P<0.05);Compared with the control group,the expression of SOX9 in the PEMF group was increased,and the difference was statistically significant(P<0.05);The expression of COL2A1 in the OA group was lower than that in the Control group,and the difference was statistically significant(P<0.05);The expression of Beclin1 in OA group was lower than that of the Control group,and the difference was statistically significant(P<0.05).The Beclin1 of the PEMF group was higher than that of the OA group,but the difference was not statistically significant(P>0.05).The expression of P62 in the control and PEMF group decreased compared to the OA group with statistical significance(P<0.05,P<0.01).Conclusion: Pulse electromagnetic fields may modify cartilage autophagy and serum inflammatory level by regulating m TOR signal pathway.PEMFs inhibit cartilage degeneration and delay progression of knee osteoarthritis in rats. |
