| Objective:To investigate the role and clinical significance of Septin-9 and HIF-1α genes in the pathogenesis of cervical cancer by studying the expression of Septin-9 and HIF-1α in cervical tissues,analyzing the relationship between the two and various clinicopathological features of cervical cancer and the correlation between the expression of Septin-9 and HIF-1α in cervical cancer.Method:The data of 99 patients with cervical cancer admitted to the Department of Obstetrics and Gynecology of the first affiliated hospital of Nan Chang University from April 2020 to May 2021 were collated,and cervical cancer tissue samples of these patients were collected as experimental specimens of cervical cancer group.The information and cervical specimens of 50 patients with cervical intraepithelial lesions confirmed by pathology after colposcopic biopsy or cervical conical resection were collected as experimental specimens of the cervical lesion group,and the data and cervical tissues of 20 patients undergoing hysterectomy due to uterine fibroids,adenomyosis and other non-cervical lesions were collected as experimental specimens of the normal cervical group.Immunohistochemical and staining methods were used to detect the expression of Septin-9 and HIF-1α in the three groups of tissues(169 samples),and their correlation with clinical indicators(age,clinical stage,pathological grade,pathological classification,lymph node metastasis and depth of muscle infiltration)was analyzed.Statistical methods were used to compare and analyze the differential expression of different types of cervical tissues.Results:1.Through univariate analysis of the general data of cervical lesions and normal tissues,it was concluded that there were statistically significant differences in age,HPV,HPV16/18,menopause and pregnancy times among the tissue types of cervical lesions(P<0.05),there was no statistically significant difference between yields(P>0.05);2.The expressions of Septin-9 and HIF-1α in cervical cancer tissues were significantly higher than those in cervical intraepithelial neoplasia group,and the expressions of Septin-9 and HIF-1α in cervical intraepithelial neoplasia group were significantly higher than those in normal tissue group(P<0.05);3.There were significant differences between the expression intensity of Septin-9and HIF-1α in cervical cancer group and clinical stage,lymph node metastasis and muscle infiltration(P<0.05),there were no significant differences in age,pathological grade and pathological classification(P>0.05);4.There was a positive correlation between the expression of Septin-9 and HIF-1α in cervical cancer tissues(r=0.671,p<0.05).Conclusion:1.The expression levels of Septin-9 and HIF-1αin cervical lesions were significantly higher than those in normal cervical tissues and gradually increased with the increase of lesion grade,which may be the pathogenic genes leading to cervical cancer.2.The expression of Septin-9 and HIF-1αin cervical cancer tissues was not correlated with age,pathological grade and pathological type.It is closely related to tumor staging,lymph node metastasis and muscle infiltration,which is of great significance for the diagnosis and treatment of cervical cancer. |
