Study On The Molecular Mechanism Of LINC00882 Promoting The Occurrence And Development Of Malignant Glioma

Background:Glioma is one of the common primary malignant brain tumors with low survival rate.More and more related literatures have reported that long-chain non-coding RNA(LncRNA)can participate in a variety of biological processes of related tumors.Objective:The role of LINC00882 in glioma has not been discovered and studied.This paper mainly studies the potential significance and mechanism of LINC00882 in the malignant phenotype of gliomas,and provides a certain value for finding potential targets for the treatment of gliomas.Methods:One of the differential genes LINC00882 was screened by RNA-seq sequencing of glioma tissue samples and verified by RT-qPCR in HEB,U87,T98G,U251 and U118 cell lines.The hsa-miR-30a-3p related to LINC00882 was found by bioinformatics analysis.T98G and U87 cell lines with significant differences in expression were selected for functional phenotypic experiments.To explore its molecular mechanism in the development of glioma.Results:The expression of LINC00882 in HEB,U87,T98G,U251 and U118 cell lines was verified by RT-qPCR.The results showed that T98G and U87 cells were highly expressed,so T98G and U87 cells were used for related experiments.After knocking down LINC00882,it was found that the ability of invasion,migration and proliferation of cells decreased by Transwell,cell scratch and CCK8 experiments,but the result was opposite after overexpression of LINC00882.After mimics,hsa-miR-30a-3p gene was tested by Transwell,cell scratch and CCK8.The results showed that it inhibited the invasion and migration of glioma cells and weakened its ability of proliferation.Conclusion:1.LINC00882 is highly expressed in glioma cells T98G and U87.2.Knockdown or overexpression of LINC00882 can inhibit or promote the invasion,migration and proliferation of glioma cells respectively.LINC00882 is the oncogene of glioma.3.The expression of hsa-miR-30a-3p in T98G and U87 increased after knocking down LINC00882.4.Mimics of hsa-miR-30a-3p gene can inhibit the invasion,migration and proliferation of glioma cells.
