Objective:To investigate the expression of S100A11 protein in intrahepatic cholangiocarcinoma(ICC)tissues and its correlation with clinical pathological factors and patient prognosis.Methods:Seventy-one cancer histopathological sections and twenty paracancer histopathological sections of patients who underwent radical surgery and had postoperative pathological confirmation of intrahepatic cholangiocarcinoma at the Department of Hepatobiliary and Pancreatic Surgery II,First Hospital of Jilin University were collected from September 2018 to December 2021.Immunohistochemistry revealed the presence of S100A11 in both intrahepatic cholangiocarcinoma and paraneoplastic tissues.The clinical data of the patients were collected,including gender,age,surgical method,whether cirrhosis was combined,preoperative CEA level,preoperative CA19-9 level,lymph node metastasis,TNM stage,depth of tumour infiltration,degree of differentiation,tumour size,nerve infiltration,vascular infiltration,liver function grading,as well as the time of tumour recurrence and overall survival time after surgery.SPSS 25.0 statistical software was employed to analyze the data,with chi-square and Fisher’s exact tests being used to assess the correlation between S100A11 expression and clinicopathological features.KaplanMeier was utilized to illustrate survival curves,and log-rank test(log-rank test)was employed to perform univariate screening.Statistically significant factors were analysed using cox regression models as independent risk factors for RFS and OS in ICC patients.Results:1.S100A11 is highly expressed in ICC tumour tissues.By analysing the TCGA database and the GEO dataset,it was found that S100A11 was significantly upregulated in ICC tissues compared to normal tissues.Immunohistochemical results showed that S100A11 was highly expressed in 63.38% of intrahepatic cholangiocarcinoma tissues and only 15.00% of paraneoplastic tissues.the difference in expression of S100A11 between intrahepatic cholangiocarcinoma tissues and paraneoplastic tissues was statistically significant(P<0.05).2.The correlation between S100A11 protein expression and clinical factors of ICC patients was analysed using chi-square test and Fisher’s precision probability test.The results showed that high S100A11 expression was associated with preoperative CEA level,preoperative CA199 level,lymph node metastasis,TNM stage,Ki-67 level and tumour vascular infiltration with significant differences(P<0.05).While there was no significant difference with patient age,gender,whether combined with cirrhosis,tumor diameter,surgical approach,degree of differentiation,tumor size,nerve infiltration,vascular infiltration and liver function grading(P>0.05).In 71 ICC patients,high S100A11 expression predicted later N and TNM staging,higher preoperative levels of CEA and CA199,higher levels of Ki-67,and a higher likelihood of vascular infiltration.3.Kaplan-Meier univariate analysis showed that S100A11 expression levels preoperative CEA,preoperative CA19-9,lymph node metastasis,TNM stage,tumour number,vascular infiltration and Ki-67 expression levels were unfavourable prognostic factors for patients’ RFS and OS.4.COX regression analysis showed that high S100A11 expression,preoperative CEA ≥5ng/m L were independent prognostic risk factors for RFS in ICC patients.high S100A11 expression,preoperative CEA ≥5ng/m L and lymph node metastasis were independent prognostic risk factors for OS in ICC patients.Conclusions:1.In intrahepatic cholangiocarcinoma tissues,S100A11 was highly expressed,and its expression was associated with preoperative blood CEA and CA19-9 levels,lymph node metastasis,TNM stage,tumor vascular infiltration,and Ki-67 levels in patients.2.Patients undergoing ICC surgery were independent risk factors for overall survival,such as high S100A11 expression,preoperative blood CEA greater than5ng/m L,and lymph node metastasis.3.High S100A11 expression and preoperative blood CEA ≥5ng/m L were independent risk factors for recurrence-free survival in patients undergoing ICC surgery.
