| Objective:The present study aims to investigate the profiling and functional prediction of urinary exosomal lncRNA of idiopathic membranous nephropathy(IMN)patients,and to explore its possible mechanism of action in IMN.Method:Sequencing data of urinary exosomal lncRNA from five IMN samples and five healthy control(HC)samples were downloaded from the NCBISRA database,and bioinformatics analysis was applied to cluster the differentially expressed lncRNA.Gene Ontology(GO)analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were used to predict the functions and possible signaling pathways involved in the differentially expressed lncRNA in IMN.Result:A total of 14865 up-regulated lncRNA and 22819 down-regulated lncRNA were detected between the IMN and HC groups.Using Fold change ≥ 2 and P value ≤ 0.05 as screening criteria,353 lncRNA were differentially expressed,18 up-regulated lncRNA and 335 down-regulated lncRNA,respectively.Most of the differentially expressed lncRNA were less than 500 nt in length,distributed in autosomes,and mostly originated from intergenic regions.GO analysis suggested that the up-regulated lncRNA might be involved in mesenchymal cell apoptosis,nitric oxide synthase activity regulation,hormone metabolism regulation,and heat shock protein 90 binding;the down-regulated lncRNA might be involved in interleukin-10 secretion.KEGG analysis suggested that the up-regulated lncRNA might be involved in m TOR signaling pathway,HIF-1 signaling pathway;the down-regulated lncRNA might be involved in Rap1 signaling pathway.Conclusion:1.The study revealed the expression and functional profile of differentially expressed urinary exosome lncRNA in IMN patients.2.Differentially expressed lncRNA in urinary exosomes may be involved in the development of IMN through mTOR signaling pathway,HIF-1 signaling pathway,Rap1 signaling pathway and other pathways. |
