Background and Purpose:The gallbladder is interpreted as a malignant tumor,which is commonly found in the epithelium of the gallbladder in the biliary system.Disease early diagnosis,found that the probability is low,the course fast,transfer time,early prognosis is poor.Among them,the surgical resection is considered to be the most effective treatment method,but the probability of successful operation excision is low.Chemotherapy is less effective,and radiation is less effective.Therefore,to establish system timely and effective treatment,it is very important to improve the effect of the treatment of gallbladder cancer.The research based on cell level and animal model is an important stage of cancer prevention and treatment.Whether tumor pathogenesis research,or tumor treatment research of new drugs,or research and exploration of new cancer therapies,and related animal models are inseparable.Therefore,we established a subcutaneous xenograft tumor model of gallbladder cancer by subcutaneous transplantation of human gallbladder cancer cells in nude mice,discussed the disease,observed the infiltration and metastasis of cancer,compared the advantages and disadvantages of various subcutaneous transplantation tumor models,and discussed the advantages and disadvantages of different gallbladder cancer cell lines in animal models,as well as the more suitable for subcutaneous transplantation of gallbladder cancer cell lines.method:The expression and location of GBC-SD,NOZ and SGC gallbladder cancer cells in the immunofluorescence assay were determined by establishing a mouse animal model,subcutaneously transplanting and injecting gallbladder cancer cell lines,and the protein expression and location of VEGF,LYVE-1,PD-L1,three different factors and receptors in tumor tissue and its metastatic lymph nodes were also detected,and statistical analysis was carried out on this basis.The differences in behavior,tumor morphology,tumor histopathology and tumor molecular pathology of three gallbladder cancer cell lines were observed and discussed.result:The average optical density of the above experimental results showed that the expression of PD-L1,LYVE-1 and VEGF proteins were different in gallbladder carcinoma tissues,and the expression level of tumor in the inguinal region of mice inoculated with gallbladder carcinoma cells was significantly different.The results of PD-L1(F=8.746,P=0.002)confirmed that the expression of GBC was the highest among the three gallbladder carcinoma cell lines,while the expression of SGC was the lowest.In the results of LYVE-1(F=6.127,P=0.010),the expression of SGC was lower than that of GBC and NOZ.Compared with SGC,the difference between GBC and NOZ was almost negligible.The results of VEGF expression(F=0.072,P=0.986)confirmed that there was no significant difference between the three gallbladder cancer cell lines;The experimental results showed that GBC and NOZ were stronger in growth and diffusion among the three different cell lines,while SGC was not prominent in this aspect.Among the three different gallbladder cancer cell lines,the mice injected with GBC gallbladder cancer suspension were the most obvious.The proportion of successful tumor formation,the tumor volume formed at the injection site was the highest and the largest,and the tumor formation time was the shortest,followed by NOZ and SGC respectively.After anatomical observation of mice inoculated with cell suspension,it was found that lymph node metastasis was found only in mice inoculated with GBC and NOZ cell suspension which had formed tumors,while no similar situation was found in mice inoculated with NOZ gallbladder cancer cell suspension.Conclusion:1.Through multiple comparisons of three gallbladder cancer cell lines modeling,GBC-SD gallbladder cancer cell line has more significant results than other experimental cell lines in terms of cell proliferation and invasion,tumor growth time,lymphatic metastasis rates.It may be the most consistent gallbladder cancer cell line,which can form relatively complete tumors for observation and experiment of animal models.At the same time,among the three targets of VEGF,LYVE-1 and PD-L1,the protein expression level of GBC-SD is the most significant,which one can be used to research the gallbladder cancer-related system and hinder the proliferation and metastasis of lymph nodes.The expression result of NOZ takes the second place and can be used as an alternative,while the expression level of SGC is the worst.Its role in animal models needs further study to explore whether SGC has another application effect.2.The experiment showed that the volume and weight of tumor were observably associated with the level of malignancy.
