Objective:Bioinformatics methods were used to screen out genes that may be related to the prognosis of epithelial ovarian cancer,and the expression of these genes was detected in epithelial ovarian cancer tissues and normal ovarian tissues of different pathological stages to explore the relationship between these genes and prognosis of epithelial ovarian cancer,providing a basis for subsequent mechanism research.Methods:1.Three gene chips,GSE18520,GSE27651,and GSE54388,were selected from the public database NCBI-GEO,and gene expression differences were detected using GEO2 R.Gene Ontology(GO),Kyoto Encyclopedia of Genes and Genomes(KEGG)The visualized and Integrated Discovery Database(DAVID)were used to analyze the function of differentially expressed genes(DEGs)and evaluated their protein-protein interaction(PPI)information by using the online tool STRING.Finally,Kaplan Meier Plotter was used to evaluate the impact of key genes on prognosis.Based on the results of the above bioinformatics methods,several genes that play key roles in the prognosis of epithelial ovarian cancer were screened out.2.30 normal ovarian tissue sections with complete clinical data and90 epithelial ovarian cancer tissue sections were selected,and the expression of CENPU protein was detected by immunohistochemistry.10 normal ovarian tissue sections with complete clinical data and 24 epithelial ovarian cancer tissue sections were selected.The difference in CENPU expression between epithelial ovarian cancer and normal ovarian tissue was further verified by real-time fluorescence quantitative polymerase chain reaction and western blot.Results:1.There were 113 differentially upregulated genes in three different microarray datasets.These genes were mainly concentrated in the mitotic cycle,chromosome aggregation,nuclear division during meiosis,the establishment of chromosome localization,and regulation of spindle microtubule and centromeric attachment.Through PPI analysis,36 key genes were screened out of 113 differentially upregulated genes.Through prognostic correlation analysis,14 genes with high expression were screened out from 36 genes that were significantly correlated with the prognosis of epithelial ovarian cancer.Finally,based on the above combination,the centromere-related gene CENPU was selected for further research.2.CENPU protein expression was significantly up-regulated in epithelial ovarian cancer tissues,and the difference was statistically significant between CENPU protein expression in epithelial ovarian cancer tissues and normal tissues.Conclusion:1.High expression of genes CENPF,CENPF,and CENPU,which encode centromere protein,were significantly associated with the poor prognosis of epithelial ovarian cancer.2.The expression of CENPU,a member of the centromere protein family,was significantly up-regulated in epithelial ovarian cancer.
