| 1.Objective(1)Analyze the research status,hotspot research and development trend of Banxia Houpu Decoction based on the visualization map;(2)To explore the curative effect and mechanism of Jinsangliyan Capsules by establishing a chronic pharyngitis rat model;(3)Analyze the relationship between the content of T cell subsets(CD3,CD4,CD8)in the blood of rats with chronic pharyngitis,the ratio of CD4/CD8 and the expression level of PGE2 in the pharyngeal tissue and chronic pharyngitis,and further determine the relationship between Jinsangliyan Capsules Mechanism of action in the treatment of chronic pharyngitis.2.Method2.1 Literature researchTaking "Banxia Houpu Decoction" as the key word,the China National Knowledge Infrastructure(CNKI)database was searched,and the time period was from the establishment of the database to December 2022.Cite Space6.1.R6 software and VOSviewer software were used for visual knowledge map analysis.2.2 Animal experimentsAfter adaptively feeding 48 healthy SPF SD rats for 1 week,they were randomly divided into 2 groups,8 in the normal group and 40 in the experimental group(with 8 as a unit),and fed in separate cages.The normal group was fed for 30 days without any treatment;the experimental group was sprayed with 2.5% ammonia water every day using a throat sprayer for 30 consecutive days,at 9 o’clock in the morning and at 3o’clock in the afternoon,every day at 9 o’clock in the morning and 3 o’clock in the afternoon.Press to make a chronic pharyngitis rat model,and judge whether the model is successful by observing the symptoms and signs of the two groups of rats.After successful modeling,the remaining rats in the experimental group were randomly divided into 5 groups,7 rats in each group,which were the model control group,the cediiodine buccal tablet group,and the low-dose group,medium-dose group,and high-dose groups of Jinsangliyan Capsules.Both the normal group and the model control group were treated by intragastric administration of the same amount of normal saline.The low,medium and high dose groups of Jinsangliyan Capsules were treated by intragastric administration of different doses of Jinsangliyan Capsules aqueous solution.The aqueous solution of cediodine buccal tablets was used for intragastric administration,and the administration was administered by intragastric administration for 15 days,twice a day.15 days later,6 rats in each group were sacrificed after anesthesia,and the required specimens were collected,and HE staining was performed on the relevant tissues of the pharynx for comparative observation,and flow cytometry was used to detect T cell subsets(CD3,CD4,CD8,CD4/CD8),record the data,use the ELISA method to measure the content of PGE2 in the pharyngeal tissue,and record the value.The recorded data were imported into SPSS26.0 statistical software for statistical analysis.3 ResultsIn the visual map analysis,a total of 1031 documents were finally included,and the annual volume of published papers showed a fluctuating upward trend;the main contributors to the cooperation are represented by Kong Lingdong and Tong Yao;the research institutions are mainly Chinese medicine universities and Chinese medicine hospitals;the high-frequency keywords are Meiheqi,chronic pharyngitis,Jingfang,etc.Hot spots and future development trends may be mutual(or multiple)research among10 clusters such as #0 Meiheqi,#1 Jingfang,and #2 chronic pharyngitis.In the animal experiment part,the contents of PGE2 in the normal group,model control group,high-dose group,middle-dose group,low-dose group,and cediiodine buccal tablet group of Jinsangliyan Capsules were 26.95±2.88,132.43±5.68,61.95,respectively.±4.98,37.74±5.01,99.39±6.56,35.67±3.37.This shows that Jinsangliyan Capsules can reduce the PGE2 content in rats with chronic pharyngitis.The CD3 contents in the normal group,the model control group,the high-dose group,the middle-dose group,the low-dose group of Jinsangliyan Capsules,and the cediiodine buccal tablet group were 52.12±1.85,33.43±7.31,46.77±2.27,and 32.10,respectively.±4.96,41.07±3.96,44.22±9.44;CD4 contents were 43.83±1.94,64.50±2.13,47.50±0.93,62.73±1.70,54.58±4.06,46.62±2.02;CD8 contents were 54.65±2.13,32.83 ±3.51,51.13±0.68,34.60±1.29,37.88±6.28,51.27±2.07;CD4/CD8 ratios were0.80±0.07,1.99±0.26,0.93±0.03,1.82±0.11,1.47±0.24,0.91±0.07,indicating Jinsangliyan Capsule has the effect of increasing the CD3 and CD8 content in rats with chronic pharyngitis,and can also decrease the CD4 content and CD4/CD8 ratio.4 ConclusionThrough the analysis of visual knowledge map,it can be seen that in the research process of Banxia Houpu Decoction,the clinical application of famous doctors’ experience,data mining of classic prescription theory and discussion of animal experiment mechanism may be the hot research directions in the future.Pharyngitis is the main keyword.This study closely followed the research hotspots,and explored the treatment and possible mechanism of Jinsangliyan Capsules based on Banxia Houpu Decoction on chronic pharyngitis through animal experiments.According to the experimental results,it was found that Jinsangliyan Capsules can not only effectively reduce the expression level of PGE2 in the pharyngeal tissue of rats with chronic pharyngitis,but also reduce the expression level of CD4 in the blood and increase the expression levels of CD3 and CD8.Therefore,Jinsangliyan Capsule may treat chronic pharyngitis by reducing the expression level of PGE2 in pharyngeal tissue,reducing the expression level of CD4 in blood,and increasing the expression level of CD3 and CD8.It can be seen from the ratio of CD4/CD8 that the therapeutic effect of the high-dose group is similar to that of the cediiodine buccal tablet group,and the effect is equivalent,and the effect after treatment is close to that of the normal group.It can be inferred that Jinsangliyan Capsules may treat chronic pharyngitis by anti-inflammation and correcting immune dysfunction. |
