| Objective:The expression levels of NLRC3 and FOXP3 in normal endometrium and ovarian ectopic endometrium of patients with endometriosis(endometriosis,EMT)were detected and compared from m RNA and protein levels by immunohistochemistry,Western Blot and RT-q PCR.In endometriosis of different stages,the difference of expression of NLRC3 and FOXP3 were explored,and the relationship between NLRC3,FOXP3 and clinicaopathological features of EMT was analyzed.Thus,the roles of NLRC3 and FOXP3 in the occurrence and development of EMT were speculated.Methods:A total of 25 patients with normal endometrium and 40 patients with EMT of ovary were diagnosed by histopathological after operated invasive treatment in Affiliated Hospital of Chengde Medical College from November 2020 to April 2022 were collected.The data were summarized according to the stages of EMT by the American Society for Reproductive(ASRM).In total,22 cases of stage III tissue specimens and 18 cases of stage IV tissue specimens were obtained.The expression levels of NLRC3 and FOXP3 in normal endometrium and EMT patients were analyzed and processed by RT-q PCR,immunohistochemistry and Western Blot.We set out to investigate the differing of expression of NLRC3 and FOXP3 in patients with stage III and IV EMT.After that,discussed the connection between the expression of NLRC3,FOXP3 and the clinical pathological characteristics of EMT.And discussed the correlation between NLRC3 and FOXP3.Results:1.Expression of NLRC3: the results of immunohistochemical,Western Blot and RT-q PCR showed that in both the stage III and stage IV EMT groups,the expression of NLRC3 protein was lower than that in normal endometrium group,the difference was statistically significant(P < 0.0125).Between the stage III and stage IV EMT groups,the expression of NLRC3 was no significant difference(P > 0.05).2.Expression of FOXP3: the results of immunohistochemistry,Western Blot and RT-q PCR showed that compared with the normal endometrium group,the expression of FOXP3 in both the stage III and IV EMT groups was higher,and the difference was statistically significant(P<0.0125).Between the stage III and stage IV EMT groups,the expression of FOXP3 was no significant difference.(P >0.05).3.Relationship between the expression of NLRC3,FOXP3 and clinicopathological features of EMT: Analysis of pathological sections revealed a lower positive rate of NLRC3 expression in tissues with higher CA125 levels.While in tissues with higher CA125 levels,the expression of FOXP3 was higher,the difference were statistically significant(P < 0.05).what’s more,in EMT patients of different ages,different lesion sizes and different dysmenorrhea degrees,the expression level of NLRC3 and FOXP3 were not statistically significant(P > 0.05).4.Correlation analysis of expression level of NLRC3,FOXP3 in EMT:NLRC3 expression and FOXP3 expression showed a negative correlation(r=-0.613,P<0.05).Conclusions:The pathogenesis of endometriosis is related to the immune imbalance in the abdominal microenvironment,the down-regulation of immune surveillance represented by NLRC3 and the enhancement of immune escape represented by FOXP3 participated in the immune imbalance of the abdominal microenvironment.The combined detection of the expression of NLRC3,FOXP3 and CA125 indicators has high application value for the diagnosis of endometriosis.Targeted treatment of the immune microenvironment of EMT dysregulation by interfering with the expression of NLRC3 and FOXP3 may be a potential pathway for new treatments of EMT. |
