Objective: To establish a mouse model of allergy induced by Echinococcus granulosus,and to investigate the changes of Treg cells,Th17 cells and CD3CD4CD8 cells in hydatid echinococcus sensitization.Methods: Echinococcus was extracted from the liver of sheep naturally infected with Echinococcus granulosus.After 40 days of culture,the Echinococcus granulosus was cultured to a diameter of 200 to 300 microns,and then each Balb/c mouse were inoculated with 50 microcapsules by intraperitoneal injection.After infection for 6 months,mouse was sensitized by intraperitoneal injection of 0.1ml/10 g of larval Echinococcus granulosus capsule.The blank group was injected with sterile saline as control.After the stimulation of allergy for 1hour,mouse were divided into normal control group,non-sensitized group and allergy group according to the score,with 6 mouse in each group.The inner epicanthus venous blood was taken from each mouse,and the spleen was aseptically taken to prepare the splenic cell suspension.Dendritic Cell(DC),regulatory T cells(Treg),helper Cell 17(Th17),CD3 cells,CD4 cells,CD8 cells,interleukin-10(IL-10)and transforming growth factor-β1(TGF-β1)and interleukin-17A(IL-17A)were assayed by flow cytometry.Results: The percentage of DC cells in the non-allergy group and the allergy group decreased significantly(P<0.001 and P<0.01,respectively.)when compared with the control group.Compared with the non-allergy group,the percentage of DC cells in the allergy group increased significantly(P<0.01).The percentage of Treg cells in the non-allergy group was significantly higher than that in the control group(P<0.01).However,there was no significant difference in the percentage of Treg cells in the allergy group compared with the control group(P>.05).Compared with the non-allergy group,the percentage of Treg cells in the allergy group decreased significantly(P<0.05).Compared with the control group,the percentage of Th17 cells in the non-allergy group was significantly lower(P < 0.05),while the percentage of Th17 cells in the allergy group was significantly higher(P < 0.01).Compared with the non-allergy group,the percentage of Th17 cells in the allergy group increased significantly(P<0.01).Compared with the control group,IL-10 levels were significantly higher in both the n non-allergy group and the allergy group(P<0.001 and P<0.05,respectively).Compared with the non-allergy group,IL-10 content in the allergy group decreased significantly(P<0.05).Compared with the control group,the content of TGF-β1 levels in both the non-allergy group and the allergy group were significantly increased,P < 0.01 and P < 0.05,respectively.Compared with the non-allergy group,TGF-β1 was significantly decreased in the allergy group(P<0.05).Compared with the control group,IL-17 A content in the non-allergy group was significantly decreased(P<0.05).Compared with the control group,IL-17 A level in the allergy group was significantly higher(P<0.001).IL-17 A was significantly higher in the allergy group than in the non-allergy group(P<0.001).Compared with the control group,the percentage of CD3 and CD4 cells in the non-sensitized group decreased significantly(P<0.05),while the percentage of CD8 cells increased significantly(P<0.01).Compared with the control group,the percentage of CD3 and CD4 cells in the sensitized group increased significantly(P<0.05 and P<0.001,respectively),while the percentage of CD8 cells did not change significantly.Compared with the non-sensitized group,the percentage of CD3 and CD4 cells in the sensitized group increased significantly(P<0.05),and the percentage of CD8 cells decreased significantly(P<0.01).Conclusion: the number of Treg cells,CD8 cells and the expression of IL-10 and TGF-β 1 in BALB/c mice infected with Echinococcus granulosus after challenged by Echinococcus granulosus crude cyst fluid decreased significantly,while the number of DC cells,Th17 cells,CD3 cells and CD4 cells increased and the content of IL-17 A increased,which eventually led to the imbalance of Th17/Treg/CD3CD4CD8 cells and the allergic reaction of Echinococcus granulosus cyst fluid overflow.
