Objective:By detecting the changes of the expression levels of inflammatory bodies(NLRC4)and cysteine aspartic specific protease 1(Caspase-1)in the serum and right atrial ear tissues of atrial fibrillation patients,the regulation and clinical significance of the two atrial fibrillation(AF)in the development of atrial fibrillation(AF).Method:Using the cross-sectional study method,200 patients with atrial fibrillation treated in our hospital from December 2020 to December 2021 were selected,among which the atrial fibrillation,there were 61 patients in the paroxysmal atrial fibrillation(PaAF)group,71 patients in the persistent atrial fibrillation(PeAF)group,and 68 patients in the permanent atrial fibrillation(PmAF)group.90 sinus rhythm(SR)patients were selected as control group.At the same time,6 patients with atrial fibrillation and 6 patients with sinus rhythm who underwent surgery in the cardiac surgery department of our hospital from December 2020 to July 2021 were selected.Intraoperative tissues of the right atrial ear were taken,and the structure and morphology of myocardial tissue and degree of myocardial fibrosis were observed by HE staining and Masson staining.The expressions of NLRC4 and Caspase-1 were detected by immunohistochemical staining.The levels of CARD-containing structural protein 4(NLRC4)and cysteine aspartate-specific protease 1(Caspase-1)in serum and PBMCs were determined by Elisa and Western blot,respectively,and the expression of pathway proteins were compared with the measured left atrial diameter(left atrial diameter)atrial diameter,LAD)for correlation analysis.Results:The results of HE staining and Masson staining showed that compared with Con group,the myocardial cell structure was destroyed and the degree of fibrosis was significantly increased in AF group.Immunohistochemical results showed that NLRC4 and Caspase-1 protein expression levels were significantly increased in AF group compared with Con group.Elisa results showed that compared with the Con group,NLRC4 and Caspase-1 expression levels in PaAF,PeAF and PmAF groups were increased(P<0.01).The NLRC4/Caspase-1 pathway protein expression levels were detected by Western blot,all in the uptrend.Pearson method was used to analyze the correlation between NLRC4,Caspase-1 and LAD.The results showed that NLRC4 concentration was positively correlated with Caspase-1 and LAD in all test groups(r=0.540,0.516,P<0.001);Caspase-1 was positively correlated with LAD(r=0.363,P<0.001).Conclusion:1.With the occurrence and progression of AF,the levels of NLRC4 and Caspase1 in patients’ serum and right ear tissues were gradually increased,suggesting that the increased expression levels of NLRC4 and Caspase-1 played a certain role in the occurrence of AF;2.As the frequency and duration of AF increased,the LAD grew larger and with the increase of LAD,the levels of NLRC4 and Caspase-1 in serum and right auricle tissue also increased3.With the occurrence and progression of AF,CRP levels in patients increased,suggesting that inflammation levels play an important role in the occurrence and development of AF.
