Objective: To explore the changes and characteristics of intestinal flora in colon cancer patients of Li and Han nationalities in Hainan.Methods: 102 patients with colorectal cancer who were treated in our hospital from October 2021 to September 2022 were selected and divided into Li group(42 cases)and Han group(60 cases)according to different nationalities.The general clinical data,surgical and pathological examination results and stool of the two groups of patients were collected.Through stool bacterial culture,the distribution of intestinal flora of the two groups was analyzed by high-throughput sequencing,and the changes and characteristics of their intestinal flora were compared.Results: The results of high-throughput sequencing analysis showed that the total sequence numbers of Han and Li groups were 61056 and 85228,respectively,and the total OTUs were 1362 and 1685,respectively.The number of OUT increased with the increase of sequencing quantity.The Shannon Wiener curve was gradually flat,and the Shannon index was gradually saturated,indicating that the sequencing depth was reasonable and the sequencing data was large.Among them,10 bacterial phyla and110 bacterial genera were identified by high-throughput sequencing analysis.The diversity index of flora in the Han group and Li group was(4.86 ± 0.58),(5.13 ±0.54),and the abundance of flora was(278.63 ±19.15),(292.52 ± 20.86),respectively.The Li group was significantly higher than the Han group,with statistical difference(P<0.05),and the abundance of Bacteroides,Firmicutes,Fusobacteria,and Proteus in the two groups were higher.The two groups of bacteria genera with higher abundance include Bacteroides,Enterobacteriaceae,Fusobacterium,Clostridium,Proctor and Megacoccus;According to the results of fecal bacterial culture,the abundance of intestinal bacteria in the two groups of patients was high,including Firmicutes,Bacteroides,Proteus and Fusobacteria.The levels of intestinal bacteria in the Han group and Li group were(25.42 ± 1.26),(56.78 ± 3.44),(8.31 ± 1.06),(2.24 ± 0.98)and(24.97 ± 1.29),(59.12 ± 4.63),(7.94 ± 0.97),(1.96 ± 0.85),respectively There is no significant difference between Proteobacteria and Fusobacteria(χ 2 were 1.758,1.796 and 1.498 respectively(P>0.05).However,Bacteroides in the Li nationality group was significantly higher than that in the Han nationality group(χ2=2.929,P<0.05);The results of high-throughput sequencing showed that the levels of Enterobacteriaceae,Clostridium,Fusobacterium and other intestinal bacteria in Han and Li groups were(1.56 ± 0.17),(2.62 ± 0.53),(3.98 ± 0.71)and(1.53 ± 0.15),(2.49± 0.61),(4.14 ± 0.69)respectively,and there was no significant difference between the two groups(χ 2 were 0.920,1.145and1.133 respectively(P>0.05).Bacteroides and Pullorum in Han group and Li group were(13.68±1.31)、(1.43 ± 0.26)and(14.31±1.27)、(1.57 ± 0.29)respectively.Li group was higher than Han group,and the difference was statistically significant(χ 2=2.420,2.552,both P<0.05);There was no significant difference in the relative gene expression between the Han nationality group and the Li nationality group(2.16 ± 0.28),(5.39 ± 1.31),(3.52 ± 0.92),(2.21± 0.23),(5.44 ± 1.28)and(3.63 ± 0.84)(χ 2 were 0.953,0.191,0.616 respectively,P>0.05).The relative expression of Bacteroides fragilis gene in Li nationality group(7.89±1.45)was significantly higher than that in Li nationality group(7.27 ± 1.56).The difference was statistically significant(χ2=2.060,P<0.05).Conclusion: Bacteroides is the highest in the intestinal flora of CRC patients in Li and Han nationalities,followed by Firmicus,Proteus,Fusobacterium,Proctor and Megacoccus.The imbalance of flora exists in both groups,and the lactic acid bacteria in Li nationality group is higher than that in Han nationality group,while the content of enterobacter is lower than that in Han nationality group,indicating that the imbalance of intestinal flora may be an influencing factor to promote the occurrence and development of CRC...
