| Objective: To use data mining method to summarize the medication rules and core prescriptions of Professor Liu Tiejun in the treatment of primary biliary cholangitis(PBC),and use network pharmacology technology to carry out mechanism and verification of the core prescriptions,in order to provide theoretical basis for the transmission of the experience of famous old Chinese medicine and provide reference for clinical treatment.method:Study 1: Using data mining and other methods,sort out and analyze Professor Liu Tiejun’s cases of PBC.Establish a database based on the "auxiliary platform for traditional Chinese medicine inheritance(V2.5)".By using data mining methods such as association rules and cluster analysis,we systematically summarized Professor Liu Tiejun’s treatment methods and concepts,and obtained the medication rules and core prescriptions.Study 2: On the basis of data mining,use network pharmacological analysis to screen the chemical components and potential action targets of PBC core drug combinations based on TCMSP database,and screen PBC disease targets in OMIM and Gene Cards database.Screening the intersection gene of the target corresponding to the chemical composition of the drug combination and the PBC-related target,importing the STRING database for protein interaction network analysis,screening its core target,and using the DAVID database to annotate the intersection gene with R language for gene ontology(GO)annotation and the Kyoto Encyclopedia of Genes and Genome(KEGG)pathway enrichment.result:Study 1: A total of 109 cases were included in this study,including 12 males and 97 females.164 drugs were used,of which the frequency of cold drugs was the highest,and then warm drugs.The main flavor is sweet and bitter.The frequency of application of drugs for regulating the spleen,liver and stomach meridians was the highest;The results of association rules show that the core drug combination "Bupleurum chinense-Angelica sinensis-Paeonia lactiflora-Rehmannia glutinosa-Glycyrrhiza uralensis" is obtained under the condition of "the number of support is 30 and the confidence is 0.9".Study 2: 143 chemical components of the core drug combination of "Bupleurum chinense-Angelica sinensis-Paeonia lactiflora-Radix rehmanniae-Glycyrrhizae" were screened,655 potential targets were obtained,506 PBC disease targets were screened,and87 drug disease intersection genes were selected,including STAT3,TNF,CTNNB1,IL6,MMP9,VEGFA,AKT1,etc.Through gene action mechanism analysis and GO enrichment analysis,the GO entries obtained(P<0.05)showed a total of 304 results enrichment,including 235 biological processes,29 cell components and 40 molecular functions.The KEGG pathway was enriched to obtain 133 pathways(P<0.05)for the treatment of PBC intersection target with core drug combination.Conclusions:Study 1: Professor Liu Tiejun used Chinese medicine to treat PBC with both attack and tonic,damp and heat,and liver and spleen;Most of them are bitter,cold,warm and sweet drugs,which can be used both cold and warm,moderate and moderate,and cool and hot;Sweet and moist,slow and urgent,bitter and cold,attack and supplement at the same time.Taking the theory of Zang Du Fu Jie as the starting point,and using data mining and network pharmacology technology,the author systematically studied the drug use law and mechanism of the tutor in the treatment of PBC,with a view to providing an example for the clinical application of Zang Du Fu Jie theory in the treatment of PBC.Study 2: Using the network pharmacology technology to explore that the combination of core drugs in the treatment of PBC may work together through multiple mechanisms such as inflammation and immunity,cell proliferation and apoptosis,and cancer-related.Through the analysis of its mechanism of action,the core drug combination treatment of PBC was clarified from the perspective of modern pharmacology. |
