| Objective: Licorice,Scutellaria baicalensis,Coptidis rhizoma,Pinellia ternata,Dried ginger,and Jujube are the six herbs that make up the Gancao Xiexin Decoction,which is a remedy for typhoid fever that dates back to the Eastern Han Dynasty.The preparation method of the substance benchmark was reduced by looking at the prescription dosage,herbal base material,herbal concoction,and decoction process of Gancao Xiexin Decoction.A preliminary study on the substance benchmark’s quality standard was then conducted to explore the quantityvalue transfer relationship,laying the groundwork for the modernization study of the compound preparation of the traditional Gancao Xiexin Decoction recipe.Methods: By reviewing the relevant literature and referring to relevant books,the herbal base material,herbal concoction specifications and prescription dosage of Gancao Xiexin Decoction were determined.The preparation method for the substance benchmark of Gancao Xiexin Decoction in the Eastern Han Dynasty was recreated on the basis of adhering to the past while incorporating modern technology,and the aqueous decoction was later dried by vacuum freeze-drying to obtain the lyophilized powder of the substance benchmark of Gancao Xiexin Decoction.Thin layer chromatography and High Performance Liquid Chromatography(HPLC)were used to establish methods for determining Liquiritin,glycyrrhetinic acid,baicalin,berberine hydrochloride,epiberberine,coptisine chloride,and palmatine chloride,as well as to study quantitative transfer relationships and establish quality standards for the substance benchmarks.The characteristic peaks of six medicinal materials in Gancao Xiexin Decoction were identified by fingerprint method,and the similarity of 15 batches of material reference was calculated.Results: The traditional recipe Gancao Xiexin Decoction originated in Zhang Zhongjing’s Treatise on Cold Injuries,which was authored in the Eastern Han Dynasty.The decoction’s medicinal ingredients and its decoction-piece specifications complied with pharmacopoeia sources and requirements.The final prescription dosage of Gancao Xiexin Decoction was established by consulting ancient literature,the Development Guidelines of Classic Famous Recipes,the textbook of the 13 th Five-Year Plan "Prescription," and current clinical dosage.Jujube(12g),Pinellia ternata(12g),Licorice(12g),Scutellaria baicalensis(9g),Dried Ginger(9g),and Coptidis rhizome(3g).Reference preparation method of reducing substance: The haplx prescription medicinal materials were weighed,added 2000 ml water(additional 160 ml water absorption),soaked for 30 min,decocted to 1200 ml,filtered with a 100-mesh silk cloth,decocted to 600 ml filtrate,frozen at-80℃ for 24 h,and then vacuum freeze-dried for 72 h,to prepare Gancao Xiexin Decoction substance reference freeze-dried powder.The thin-layer identification technique was established for the six medicinal materials in the prescription,and the Gancao Xiexin Decoction reference material displayed the same color spots in the same place as the control medicinal materials,with no interference.A reliable method for determining drug reference content was developed,as well as a fingerprint of substance reference.The similarity of fingerprint of 15 batches of Gancao Xiexin Decoction was greater than 0.90.The quantitative transfer relationship was as follows: the transfer rates of glycyrrhizin,glycyrrhizic acid,baicalin,epigberine,coptiline,palmatine and berberine hydrochloride were all 49.08%,42.24%,28.34%,26.72%,29.32%,33.62% and 47.93% in the mean substance basis.Conclusion: The preparation method of Gancao Xiexin Decoction in the Eastern Han Dynasty was restored through the research of ancient classics and literature to determine the prescription dose,the primitive of medicinal materials and the processing of medicinal materials.The quality control standard of the material standard of Gancao Xiexin Decoction was initially established,and the quantity and quality transfer rule from the material standard to the decoction piece to the material standard was clarified.It provides reference and scientific basis for further study of the classic prescription Gancao Xiexin Decoction. |
