Objective : To explore T cells in lung adenocarcinoma patients serum immunoglobul in and mucin domain structure 1(Tim-1)Concentration change and the expression of the relationship between clinical parameters,TIM-1 the expression of influence on the prognosis of lung adenocarcinoma and its related mechanism,And to find a simple,can predict the prognosis of lung adenocarcinoma.Methods:1.Clinical control experiment method:Collect between January 2020 and December 2021,Dali university first affiliated hospital of 73 patients with lung adenocarcinoma and 57 healthy subjects in the Physical Examination Center,3 ml of venous blood was collected and the related clinical information collection,Using ELISA experimental determination of TIM-1 in lung adenocarcinoma patients serum concentrations,and then with healthy check-up than TIM-1 concentration in human serum,And TIM-1 concentration in serum of lung adenocarcinoma patients compared with relevant clinical parameters,TIM-1 in serum concentration changes in patients with clear levels associated with lung adenocarcinoma,the relationship of clinical parameters,TIM-1 in serum by ROC curve analysis sensitivity and specificity in lung adenocarcinoma.2.Bioinformatics analysis:(1)Use the TIMER database analysis TIM-1 gene expression in different tumor types and TIM-1 in lung adenocarcinoma tissues and normal tissues;(2)Using GEPIA database analysis TIM-1 gene expression level in lung adenocarcinoma tissue’s influence on the prognosis of lung adenocarcinoma;(3)Using Kaplan Meier-plotter database analysis TIM-1 gene expression level in lung adenocarcinoma tissue’s influence on the prognosis of lung adenocarcinoma;(4)Use TIMER database analysis TIM-1 gene expression and correlation of immune cell infiltration;(5)Using the method of gene enrichment analysis(GSEA)to analyze the enrichment of TIM-1 gene signaling pathway,It is inferred that the expression of the TIM-1 gene may affect the prognosis of patients with lung adenocarcinoma through the regulation of certain signaling pathways.Results:1.Collect clinical diagnosis of lung adenocarcinoma patients serum in 73 cases,Serum levels of 57 cases of healthy check-up crowd;(1)By lung adenocarcinoma group compared with control group TIM-1 levels in serum,Show that TIM-1 concentration in serum of lung adenocarcinoma patients(median of 136.4 pg/ml,25% to 75%:66.2 to245.7 pg/ml)Higher than the control group(median of 77.9 pg/ml,25% to 75%:40.8to 161.9 pg/ml),The more similar between the two groups(P < 0.05).(2)TIM-1 in serum in the diagnosis of lung adenocarcinoma results show that the ROC curve analysis,The area under the ROC curve was 0.673(P<0.001),the cut-off value on the ROC curve was 63.6pg/ml,at this time the sensitivity was 83.6%and the Specific degrees was42.1%.TIM-1 in lung adenocarcinoma patients serum levels and the related relationship comparison of clinical parameters,TIM-1 concentration in serum of lung adenocarcinoma patients with no difference in the comparison of the patients with age(Age < 60 years old TIM-1 when the concentration of the median was 139.9pg/ml,25%to75%:71.2 to 296.9pg/ml;Age of 60 or more TIM-1 concentration of median was 154.7pg/ml,25%-75%:84.0-248.8pg/ml,P values was 0.258)TIM-1;concentration in serum of lung adenocarcinoma patients and no difference in the comparison of the patients with sex(The median concentration of male patients with TIM-1 was165.1pg/ml,25%-75%:100.3-307.4pg/ml;Women with TIM-1 concentration of median was 136.3pg/ml,25%-75%:70.0-236.6pg/ml,P values was 0.246);TIM-1concentration in serum of lung adenocarcinoma patients compared with patients with or without smoking history of no difference(TIM-1 in patients with no history of smoking concentration of median was136.2pg/ml,25%-75%:50.3-233.6pg/ml,Patients with a history of smoking,TIM-1 concentration of median was152.4pg/ml,25%-75%:91.2-299.6pg/ml;P values was 0.734);TIM-1 concentration in serum of lung adenocarcinoma patients and patients if there is no difference in the comparison of lymph node metastasis(TIM-1 patients with lymph node metastasis concentration of median was 137.7pg/ml,25%-75%:63.9-213.3pg/ml;No lymph node metastasis in patients with TIM-1 concentration of median was154.7pg/ml,25%-75%:100.8-460.3pg/ml;P values was0.082);TIM-1 concentration in serum of lung adenocarcinoma patients with different TNM staging in patients with no difference in the comparison(Patients with stage I + II of TIM-1concentration of median was135.6pg/ml,25%-75%:449.1-192.7pg/ml;Patients with stage III + IV of TIM-1concentration of median was150.1pg/ml,25%-75%:72.4-270.8pg/ml;P values was0.480).2.Bioinformatics analysis results:(1)TIM-1 gene cancer analysis:Use TIMER2.0online analytical tools to assess the TIM-1 expression in generic carcinoma,With the block diagram shows the distribution of the level of gene expression,In all 34 types of cancer,There are 10 types of cancer of the observed in tumor tissue TIM-1 high gene m RNA expression,Including bladder urothelial carcinoma,bile duct cancer,colon cancer,renal clear cell carcinoma,renal papillary carcinoma,hepatocellular carcinoma,lung adenocarcinoma,adenocarcinoma and gastric cancer,In renal cell carcinoma tissues,TIM-1 gene too low m RNA expression,This shows that TIM-1 in different cancer types have different expression patterns.(2)TIM-1 in lung adenocarcinoma and expression in normal lung tissue:Using the GEPIA2 database,Lung adenocarcinoma and TIM-1 single gene expression in normal lung tissue variance analysis,Results in lung adenocarcinoma tissues TIM-1 gene m RNA expression level is significantly higher than normal lung tissue,The difference was statistically significant,P<0.001.(3)TIM-1the expression level of relationship with the prognosis of lung adenocarcinoma:Use the Kaplan-Meier plotter database analysis of TIM-1 gene in lung adenocarcinoma of high and low expression level in patients with lung adenocarcinoma.(4)TIM-1 and immune cell correlation analysis:Use the TIMER database to explore the expression and infiltration in lung adenocarcinoma TIM-1 the relationship between the immune cells,The results show that,TIM-1 expression in lung adenocarcinoma tissue and tumor purity and B cells,CD8 + T cells and CD4 + T cells,macrophages,and neutral grain.(5)TIM-1 enrichment analysis:By using the GSEA analysis high TIM-1 gene expression in the tumor samples are enriched in complex control r RNA expression、formation of the beta catenin TCF transactivating complex、hdacs deacetylate histones、Rho gtpases activate pkns、RNA polymerase I promoter escape.Enrichment analysis showed that TIM-1 high expression may promote lung adenocarcinoma cancer cell proliferation by the biological processes and affect clinical outcomes for patients with lung adenocarcinoma.Conclusion:1.TIM-1 gene in the generic carcinoma analysis showed,Compared with the corresponding normal tissues,TIM-1 in Cholangiocarcinoma、Bladder urothelial carcinoma、Colon cancer、Renal clear cell carcinoma、Renal papillary carcinoma、HCC、Lung adenocarcinoma、Straight adenocarcinoma、Cancer of the stomach、Endometrial cancer.2.TIM-1 in lung adenocarcinoma patients serum concentration and in lung adenocarcinoma tissues were significantly higher than normal.3.TIM-1 high expression in lung adenocarcinoma is associated with poor prognosis of patients with lung adenocarcinoma.4.TIM-1 in lung adenocarcinoma tissue expression with the purity of the tumor and B cells,CD8 + T cells and CD4 + T cells,macrophages,neutrophils and dendritic cell infiltration is not relevant.
