| Objective: Coronary atherosclerotic heart disease(CHD)is a kind of heart disease caused by coronary artery stenosis,occlusion,or coronary artery spasm,which leads to myocardial ischemia,hypoxia or necrosis.It is the most common type of various organ lesions caused by Atherosclerosis(AS).Atherosclerosis refers to the chronic inflammation occurring in the arterial wall.More and more studies have confirmed that inflammation is involved in the formation of atherosclerosis.Finding new therapeutic methods to inhibit plaque progression or prevent its formation has become one of the hot spots in the cardiovascular field.Previous studies have shown that NLRP3 and Treg/Th17 balance play an independent role in the occurrence and development of atherosclerosis.At present,no study has shown whether NLRP3 can regulate the progression of atherosclerosis by regulating Treg/Th17 balance.This study explores the correlation between serum NLRP3 and Treg/Th17 balance in patients with coronary atherosclerosis,which provides new ideas for exploring the pathogenesis of atherosclerosis and provides theoretical basis for the treatment of coronary heart disease.Methods: A total of 52 patients with at least one coronary artery diameter stenosis ≥50%confirmed by coronary angiography who were hospitalized in Wuhan Central Hospital from May 2022 to February 2023 were selected as the coronary heart disease group,and 52 patients with coronary artery stenosis < 50% confirmed by coronary angiography in the same period were selected as the non-coronary heart disease group.The clinical data of the research objects were sorted out.Fasting venous blood samples were collected from the subjects for routine blood tests and related indicators were recorded.Peripheral blood mononuclear cells of the two groups were isolated and cultured.The percentages of Treg and Th17 cells were determined by flow cytometry,and the ratio of Treg/Th17 was calculated.Enzyme-linked immunosorbent assay(ELISA)was used to detect serum NLRP3,interleukin(IL)-10,transforming growth factor(TGF)-β1,Il-1β,The levels of IL-17 A and IL-23 were detected,and the results were compared.Pearson correlation analysis was used to analyze the correlation between serum NLRP3 level and Treg/Th17 ratio in CHD group.Results: Flow cytometry showed that compared with the non-CHD group,the CHD group had a significantly increased proportion of Th17 cells(P < 0.05),while the proportion of Treg cells and Treg/Th17 ratio were significantly decreased(P < 0.05).The concentrations of NLRP3,IL-1β,IL-17 A,and IL-23 in the CHD group were significantly higher than those in the non-CHD group(P<0.05),and the concentrations of IL-10 and TGF-β1 were significantly lower than those in the non-CHD group(P<0.05).In addition,Pearson correlation analysis showed that there was a negative correlation between serum NLRP3 level and Treg/Th17 ratio in CHD group(r=-0.699,P<0.05).Conclusions: The results of this study show that serum NLRP3 level is significantly increased and Treg/Th17 ratio is decreased in patients with coronary atherosclerosis,and there is a significant correlation between NLRP3 and Treg/Th17 ratio,indicating that NLRP3 may regulate the occurrence and development of atherosclerosis by affecting the balance of Treg/Th17.However,its specific mechanism needs to be further explored. |
