Effect Of Esketamine On Serum CTnI,IL-6 And TNF-α Induced By Hindlimb Ischemia-reperfusion In Rats

Objective:To study the effect of esmketamine on myocardial injury(MI)related index serum cardiac troponin I(cTnI),interleukin-6(IL-6)and tumor necrosis factor α(TNF-α)after lower limb ischemia reperfusion(LIR)in rats.To further provide experimental basis for myocardial protection during perioperative period.Methods:A rat model of Limb ischemia reperfusion injury(LIRI)was set up(ischemia for 3 hours,reperfusion for 2 hours),and 30 male SD rats were randomly divided into five groups(n=6):sham surgery group(Sham group),ischemia-reperfusion group(IR group),sham surgery+esketamine group(Sham+Esk group),pre-ischemia esketamine+ischemia-reperfusion group(IR+Esk1 group),pre-reperfusion esketamine+ischemia-reperfusion group(IR+Esk2 group).The changes of cardiac troponin I(cTnI),tumor necrosis factor α(TNF-α),interleukin-6(IL-6)and norepinephrine(NE)in serum between different groups in the experimental group were observed by enzyme-linked immunosorbent(ELISA).Results:The contents of cTnI,TNF-α,IL-6 and NE were higher in the IR group than in the Sham group.The contents of cTnI,TNF-α and IL-6 and NE were lower and higher in the Sham+Esk group than in the Sham group,the contents of cTnI,TNF-α and IL-6 were reduced in the IR+Esk1 group and the contents of NE in the IR+Esk2 group(P<0.05).Conclusion:Esmketamine reduced serum cTnI,IL-6 and TNF-α concentrations after LIR in rats,and the reduction was more significant when administered before ligation.These results suggest that esketamine may have a protective effect on MI induced by LIR in rats.
