Objective:To investigate whether the expression levels of human CCR 4-NOT transcription complex subunit 7(CNOT 7)were significantly different in hepatocellular carcinoma(HCC)tissues and their adjacent counterpart.Analyze the effects of clinical data of HCC patients such as age,stage and CNOT7 expression level on postoperative survival time of HCC patients,and establish a survival evaluation model of HCC to look for independent prognostic factors with significant effects on HCC survival.And preliminarily explore the possible mechanism of CNOT7 affecting HCC by bioinformatics.Methods:Using immunohistochemical staining(IHC)to detect the expression level of CNOT7 in the two types of tissues,and the expression level of CNOT7 in paraffin sections was analyzed using two independent sample t-test to determine whether there was a significant difference.Collect clinical data of HCC patients to study whether age,tumor differentiation degree and CNOT7 expression level can affect the long-term survival time of HCC patients,Log rank test univariate analysis of clinical data,identify the characteristics significantly associated with survival,univariate analysis of multivariate Cox proportional risk regression analysis,improve the credibility of the results,and find out the independent risk factors for the prognosis of HCC patients.Finally,the pathways related to HCC progression were selected through KEGG database,and the possible mechanism and related signaling pathway of CNOT7 in HCC were preliminarily explored.Results:Of the 15 pairs of tissues for immunohistochemistry,10 of 15 HCC tissues highly expressed CNOT7(66.7%)and only 2 of 15 adjacent tissues(13%),with significant differences between the two groups(P<0.05).Univariate analysis showed that sex,age,history of hepatitis,presence of vascular cancer thrombus,and hypoproteinemia were not significantly different from postoperative survival,however,the degree of tumor differentiation,tumor stage,single tumor and CNOT7 expression level of HCC were significantly correlated with postoperative survival.Multivariate Cox regression analysis showed that vascular cancer thrombus formation and degree of tumor differentiation were not significantly correlated,while both tumor stage and CNOT7 expression level were independent prognostic factors.The final resulting survival prediction equation X~2=19.488,V=4,and P<0.05.KEGG enrichment analysis found that CNOT7 and its regulated downstream molecules such as signal transduction and activator of transcription 1(STAT 1)were largely concentrated in the JAK/STAT pathway.Conclusions:1.The expression level of CNOT7 in HCC tumor tissues was higher than that in adjacent tissues,and the expression difference was meaningful.2.Univariate analysis showed that the degree of tumor differentiation,tumor stage,whether solitary tumor and CNOT7 expression level were associated with survival after HCC surgery,but multivariate analysis showed that only tumor stage and CNOT7expression level could be used as a prognostic indicator of HCC independence.3.CNOT7 may influence the expression of downstream targets through the JAK/STAT pathway and regulate the development of HCC.
