Study On The Expression Of MT-CO2 And The Total Antioxidant Capacity In Peripheral Blood And Kidney In Lupus Mice

Abstract/Summary:
Objective:To investigate the relationship between the m RNA expression level of mitochondrial cytochrome c oxidase subunit 2(MT-CO2)and total antioxidant capacity and systemic lupus erythematosus(SLE)by detecting the m RNA expression level of MT-CO2 and total antioxidant capacity in peripheral blood and renal tissue between lupus mice and control mice.Methods:Ten female MRL/lpr lupus mice,aged 10 weeks,were randomly selected as the experimental group.Ten female C57BL/6 mice,aged 10 weeks,were randomly selected as the control group.Whole blood was collected by removing the right eyeball of all mice after anesthesia,and the plasma was isolated for the determination of total antioxidant capacity,while the rest of the blood cells were used to extract peripheral blood mononuclear cells(PBMC),that was extracted for the extraction of total RNA.The kidneys were isolated from the mice after killed by neck removal,one portion of which was for pathological PAS staining of renal tissue fixed with formalin and the others were for total RNA extraction and total antioxidant capacity determination.Quantitative real-time polymerase chain reaction(q PCR)was used to detect the expression level of MT-CO2 m RNA in PBMC and renal tissue between lupus mice and control mice.ABTS was used to detect the total antioxidant capacity in plasma and renal tissue between lupus mice and control mice.SPSS 24.0statistical analysis software was applied to data analysis and Graph Pad Prism 8.0software to mapping.The measurement datas conform to normal distribution were expressed as mean±standard deviation(x±s)and the differences between groups were compared by independent sample t test.Meanwhile,the non-normal distribution data were expressed as median(quartile spacing),which was(M(P25,P75)and the differences between groups were compared by the non-parametric Wilcoxon rank-sum test.The correlation among the total antioxidant capacity and the expression level of MT-CO2 in peripheral blood of lupus mice,the total antioxidant capacity in plasma and the expression level of MT-CO2 in renal tissue of lupus mice were analyzed with the Spearman correlation analysis,the total antioxidant capacity and the expression level of MT-CO2 in renal tissues of lupus mice were analyzed with the Pearson correlation analysis.P<0.05 was considered statistically significant.Results:The result of pathological PAS staining in the renal tissue of lupus mouse showed type III-Ⅳpathological changes,and diffuse mesangial cells and stromal proliferation,accompanied by glomerular crescent formation.PCR test results showed that the MT-CO2 m RNA expression level of PBMC in lupus mice0.3226(0.3025,0.3577)was lower than that in control mice 0.6553(0.6170,0.8900),and the difference was statistically significant(Z=-4.157,P<0.0001),and then the MT-CO2 m RNA expression level in the renal tissue of lupus mice 0.6378±0.15928was higher than that of control mice 0.5972±0.18728,and the difference was not statistically significant(t=0.825,P=0.418).ABTS test results showed that the plasma of total antioxidant capacity of lupus mice 0.1684(0.1466,0.1881)was lower than that of control mice 0.4644(0.4233,0.4942),and the difference was statistically significant(Z=-3.780,P<0.0001),and then the total antioxidant capacity in the renal tissue of lupus mice 0.2684±0.0760 was lower than that of control mice 0.4079±0.10079,and the difference was statistically significant(t=3.494,P=0.003).The results of correlation analysis suggested that the MT-CO2 expression level of PBMC and the total antioxidant capacity of plasma of lupus mice(r=-0.552,P=0.098),the MT-CO2expression level and the total antioxidant capacity of renal tissues(r=0.31,P=0.383)of lupus mice,and the MT-CO2 expression level in renal tissue and the total antioxidant capacity of plasma(r=0.20,P=0.58)of lupus mice were not statistically significant difference in the analysis results.Conclusion:The decrease of total antioxidant capacity of peripheral blood and kidney in lupus mice may be related to the pathogenesis of SLE.The decreased MT-CO2 expression level of PBMC may be related to the pathogenesis of SLE.The correlation between the m RNA expression of MT-CO2 in renal tissue and renal damage in lupus mice need to be further verified.At present,there was no correlation between the m RNA expression level of MT-CO2 and the total antioxidant capacity in peripheral blood and renal tissue of lupus mice.
