Study On The Effect Of Silkworm-Periostracum Cicada Sicao Decoction On Psoriasis Mouse Model By Regulating TWEAK Protein

Objective : In this study,the intervention effect of silkworm Sicao Decoction on imiquimod induced psoriasis mouse model was investigated through animal experiments.By observing the effect of the formula on TWEAK and its related inflammatory factors(Tumor necrosis factor-related weak apoptosis-inducing factors,Interleukin-23 and IL-17 A,Tumor necrosis factor-α,Transforming growth factor-β),To explore the mechanism of silkworm Sicao decoction in the treatment of psoriasis,and to provide a basis for clinical application and further study of the mechanism.Methods:The mouse model of psoriasis was established by applying imiquimod cream.After weighing and skin preparation,60 Balb/c mice were divided into 6 groups,namely blank group,model group,positive drug control group,and rigidus Sicao decoction high,medium and low concentration groups,with 10 mice in each group.The modeling of the experiment was conducted simultaneously with drug intragastric administration,both once a day for 7 consecutive days.The rats in the blank group were given normal saline by gavage once a day for 7 consecutive days.In the model group,5% imiquimod cream 62.5mg and normal saline were applied to the skin preparation area on the back by gavage once a day for 7 consecutive days.In the positive drug control group,5% imiquimote cream62.5 mg+ acitretin capsule distilled water suspension was applied to the back skin preparation area by gavage once A day for 7 consecutive days.In the high concentration group,5% imiquimote cream 62.5 mg and traditional Chinese medicine suspension20mg/kg were applied to the back skin preparation area by gavage once a day for 7consecutive days.In the medium concentration group,5% imiquimote cream 62.5 mg and traditional Chinese medicine suspension 10mg/kg were applied to the back skin preparation area by gavage once a day for 7 consecutive days.In the low concentration group,5% imiquimote cream 62.5 mg and traditional Chinese medicine suspension5mg/kg were applied to the back skin preparation area by gavage once a day for 7consecutive days.After 7 days of experiment,the lesion area and severity index(PASI)of the mice in each group were counted,and the gross pictures were taken.The histopathology of the skin lesions was observed by immunofluorescence staining,and the contents of TWEAK,IL-23,IL-17 A,TNF-α,and TGF-β in the serum of peripheral blood were detected by enzyme-linked immunosorbent assay(ELISA).Results: After 7 days of continuous intervention,compared with the model group,the PASI score and pathological manifestations of skin lesions in the high,medium and low concentration groups of Silkworm-Periostracum Cicada Sicao Decoction and the positive drug control group were decreased and improved(p < 0.05).Compared with the model group,the concentration levels of inflammatory factors TWEAK,IL-17 A,IL-23,TNF-αand TGF-β in peripheral blood serum were significantly decreased(p< 0.05).Compared with the positive drug control group,the improvement of the above observation indexes of Silkworm-Periostracum Cicada Sicao Decoction was better(p < 0.05).In the comparison of intra-group observation indicators among the high,medium and low concentration groups,the medium concentration group had the best curative effect.Conclusion : The results showed that the decoction was effective in the treatment of psoriasis,and the pathological tissue and PASI scores showed that it could improve the symptoms of psoriasis in mouse models.It can be seen from the ELISA experiment that the Silkworm-Periostracum Cicada Sicao Decoction can reduce TWEAK expression in the serum of psoriasis mouse models,inhibit the production of inflammatory factors IL-17 A,IL-23,TNF-α,TGF-β,and reduce the inflammatory reaction,so it is speculated that this prescription may play an intervention role by reducing TWEAK protein.It can provide some basis for clinical treatment of psoriasis,but the specific mechanism of action still needs further study.
