| Objective: The study aims to investigate the expression of miR-126-3p and miR-210-3p in plasma exosomes of pregnant women with preeclampsia and normal pregnant women in the third trimester,and analyze their differential expression in plasma exosomes and their clinical significance in preeclampsia.Methods: All subjects were strictly screened according to unified exclusion criteria and inclusion criteria,30 pregnant women with preeclampsia who gave birth in the Affiliated Hospital of Hangzhou Normal University from January 2021 to November 2022 were selected as the experimental group(preeclampsia group),and another 30 healthy pregnant women who gave birth in the hospital during the same period were selected as the control group(normal pregnancy group).We collected general clinical data of all subjects including age,days of pregnancy,pre-pregnancy body mass index(BMI),maximum systolic blood pressure and maximum diastolic blood pressure,qualitative examination of urine protein,etc.At the same time,we collected the peripheral venous blood of pregnant women in the two groups,and the plasma was separated and collected,then the exosomes in plasma and total RNA of exosomes was extracted.Finally,relative expression levels of miR-210-3p and miR-126-3p in exosomes were detected by real-time fluorescence quantitative PCR(qRT-PCR).At the same time,the ROC curve was used to evaluate the value of the two target miRNAs in the clinical diagnosis of preeclampsia.Results: In the preliminary test,compared with 10 cases of the preeclampsia group and 10 samples from the normal pregnancy group,the relative expression level of miR-126-3p in the plasma exosomes of pregnant women in the preeclampsia group showed a downward trend,while the relative expression level of miR-210-3p in the plasma exosomes of pregnant women in the preeclampsia group showed an upward trend.Then,we further collected samples for experiments,and the results showed that compared with 20 pregnant women in the preeclampsia group and 20 pregnant women in the normal pregnancy group,the expression level of miR-126-3p in the plasma exosomes of pregnant women with preeclampsia was down-regulated,and the difference was statistically significant(p<0.05);At the same time,the results showed that the relative expression level of miR-210-3p in plasma exosomes of pregnant women in preeclampsia group showed a higher trend than that in normal pregnancy group,and the difference was statistically significant(p<0.05).According to ROC curve,both miR-126-3p and miR-210-3p in plasma exosomes have certain value in the diagnosis of preeclampsia alone,but they have limitations.Compared with the diagnosis alone,the combined application of both has better diagnostic efficacy.Conclusion:1.Compared with the normal pregnancy group,the expression level of miR-126-3p in the peripheral blood plasma exosomes of pregnant women with preeclampsia in the third trimester was lower.2.Compared with the normal pregnancy group,the expression level of miR-210-3p in the peripheral blood plasma exosomes of pregnant women with preeclampsia in the third trimester was higher.3.The miR-126-3p and miR-210-3p in plasma exosomes of pregnant women in the third trimester may have diagnostic ability for preeclampsia,and the combined application of the two indicators is better than the auxiliary diagnosis of preeclampsia alone.4.The expression level of miR-126-3p and miR-210-3p in plasma exosomes of preeclampsia patients may be related to the occurrence and development of preeclampsia. |
